摘  要:目的:观察鼠牙周炎基础治疗前后牙龈组织中层粘连蛋白(LN)的表达。方法:丝线结扎高糖软食法建立鼠牙周炎模型。去除结扎丝线停止高糖软食,局部洁刮治术,建立鼠牙周炎基础治疗模型。免疫组化法显示LN在牙龈组织中的表达。结果:LN连续线形表达于健康牙龈上皮和血管内皮下基底膜(BM);偶见周围基质LN阳性表达牙龈成纤维细胞(GFs),位于上皮钉突MB附近。LN不连续点线形表达于牙周炎牙龈上皮和血管内皮下BM,表达强度与炎症性质和迁延时间有关;周围基质LN阳性表达GFs增多,位于上皮钉突和结合上皮(JE)根端BM附近。LN连续线形表达于基础治疗后牙龈上皮和血管内皮下BM,表达强度上调;周围基质LN阳性表达GFs减少,位于上皮钉突BM附近。结论:炎症改变牙龈BM内LN表达方式和强度,刺激上皮BM附近GFs分泌LN;牙周基础治疗恢复LN在牙龈组织内的分布,强度上调。GFs源性LN可能参与牙龈上皮细胞的增殖和JE根向迁移,上皮源性LN可能影响BM的构建和稳定。Objective: To observe the expression of laminin (LN) in rat gingiva before and after initial therapy of periodontitis. Methods: Periodontitis was induced by silk thread ligation and high glucose soft diet, then was treated by supragingival and subgingival scaling after taking off silk thread ligation and ceasing high glucose soft diet. The expression of LN was detected by immunohistochemistry. Results: Normal group. LN consecutively and linearly expressed in the basement membrane( BM), occasionally in the extracellular matrix ( ECM ) of gingival fibroblasts ( GFs ). Periodontitis group : LN inconsecutively expressed in the BM, whose Intensity was relevant with inflammation nature and duration; in teh ECM GFs enhanced mostly underlying BM of epithelium rete pegs and junctional epithelium(JE) apical tip. LN positive expression increased. Initial therapy group: LN consecutively and linearly distributed in the BM. The expression of LN decreased in ECM of GFs, which mainly distributed around epithelium rete pegs. Conclusion: Inflammatory irritants alter the expression of LN in BM, and induce the GFs underlying BM to produce LN. Initial therapy recovers the distribution of LN, and up - regulates the LN expression. LN from GFs possibility participates in the proliferation and migration of epithelium. LN from epithelial cells possibility influences the construction and stabilization of BM.
