摘 要:目的探索中国老年汉族人群中CCR5和CCR2基因多态性与原发性高血压的相关性。方法入选的高血压患者(n=187)和非高血压对照者(n=98)均为中国老年汉族人群,提取外周血基因组DNA。采用PCR法检测CCR5Δ32突变,PCR-测序法检测CCR2-64I基因型。结果所有被研究者中均未发现CCR5Δ32突变基因型。高血压组中CCR2-64I杂合子59例(31.55%),纯合子16例(8.56%),CCR2-64I等位基因频率为24.3%;非高血压对照组中CCR2-64I杂合子34例(34.69%),纯合子4例(4.08%),CCR2-64I等位基因频率为21.4%。等位基因多态性在该群体中呈Hardy-Weinberg平衡分布,两组等位基因频率及基因型比较有差别,但均无统计学意义(P>0.05)。结论该研究在中国老年汉族人群中未检出CCR5Δ32突变基因型,且未证实CCR2-64I基因位点与高血压相关。Objective To explore the effect of genetic polymorphisms of CCR5 and CCR2 in essential hypertension in Han Chinese elders. Methods Genomic DNA samples were obtained from 187 patients with hypertension and 98 healthy controls.Polymerase chain reaction(PCR) and electrophoresis were performed to identify the genotype of CCR5△32.PCR and sequencing were employed to identify the genotype of CCR2-64I. Results In CCR5 gene,none allele of 32bp deletion was found in all subjects.At CCR2-64I locus,59 heterozygotes(31.55%) and 16 homozygotes(8.56%) were observed in the hypertension patients(CCR2-64I,24.3%) and 34 heterozygotes(34.69%),4 homozygotes(4.08%) in healthy controls(CCR2-64I,21.4%).The genotype distributions observed in both groups were in accordance with Hardy-Weinberg equilibrium.However no significant difference of genotypes or the allelic frequency was identified(P〉0.05).Conclusion The results suggested that the polymorphism of CCR2-64I was not associated with hypertension in Han Chinese elders.
