乳腺癌中转化生长因子TGFβ_1 mRNA及蛋白表达及意义

摘　　要：目的探讨转化生长因子TGFβ1 mRNA及其蛋白在乳腺癌中的表达及其意义。方法采用RT-PCR和免疫组织化学法分别检测96例乳腺癌新鲜组织及对应60例石蜡组织中TGFβ1 mRNA及蛋白的表达。结果乳腺癌中TGFβ1 mRNA及蛋白的高表达率分别为45.83%、43.33%,明显高于其在乳腺良性增生组织(26.67%,20%)及正常乳腺组织(9.09%,0)中的表达,差异有统计学意义(P<0.05);且在乳腺癌腋淋巴转移组TGFβ1 mRNA及其蛋白高表达率分别为64.29%(36/56)、57.14%(20/35),均明显高于腋淋巴结无转移组的20.00%(8/40)、24.00%(6/25),两组表达的差异有统计学意义(P<0.01)。结论TGFβ1 mRNA及其蛋白在乳腺癌发病发展过程中起重要促进作用,提示TGFβ1基因可能是乳腺癌的重要促淋巴转移因子,有望成为治疗的靶标及有价值的预后判断指标。Objective To investigate the expression of transforming growth factor β1 （TGFβ1） in breast cancer and its significance. Methods TGFβ1 mRNA was detected in 96 fresh tumor tissues of breast cancer using semi-quantltatlve reverse transcription polymerase chain reaction（ RT-PCR）, and TGFβ1 protein was examined in 60 corresponding formalin-fixed paraffin-embedded tissues by immunohistochemistry. Results The overexpression rates of both TGFβ1 mRNA and protein in breast cancer tissues were 45.83% ,43.33% , respectively,whlch were higher than that in innocence hyperplastic tissues of breast（ 26.67% ,20% ）and normal breast tissues（26. 67% ,20% ）with significant difference （P 〈 0. 05 ）. Furthermore, in group with axillary lymph node metastasis and group without axillary lymph node metastasis of breast cancer, the frequency of TGFβ1 mRNA and protein overexpression rates were 64.29% （36/56） ,57.14% （20/35）and 20.00% （8/40） ,24. 00% （6/25） , respectively, the differentence of which was statistically significant（ P 〈 0.01 ）. Conclusion The results suggest that the overexpression of both TGFβ1 mRNA and protein may play an important role in tumorigenesis and progression of breast cancer, and TGFβ1 gene may be one of factors accelerating lymph node metastasis of breast cancer,which may become a target of treatment and a prognostic biomarker in breast cancer.
