摘　　要：目的探讨阿尔茨海默病(Alzheimer's disease,AD)患者外周血中巨噬细胞移动抑制因子(macro-phage migration inhibitory factor,MIF)的表达水平及其与病情程度的关系。方法应用ELISA法和荧光定量PCR技术,测定64例AD患者和50名健康对照者外周血MIF的表达水平。根据简易精神状况检查量表(MMSE)评分将AD患者分为轻、中、重度3组。结果血浆MIF的浓度和外周血单个核细胞(peripheral blood mononuclear cells,PB-MC)MIF mRNA的表达水平:AD组高于对照组,差别有统计学意义(P<0.05);AD痴呆重度组分别高于轻、中度组,差别有统计学意义(P<0.05);但轻、中度组之间差别无统计学意义(P>0.05)。蛋白和核酸两个水平MIF的浓度与MMSE评分成负相关(r蛋白=-0.34,r核酸=-0.36,P<0.01)。结论MIF可能参与AD的病理过程并发挥重要作用,其水平变化与痴呆严重程度相关。Objective To investigate the expression of macrophage migration inhibitory factor（MIF） in peripheral blood mononuclear cells （PBMC） of patients with Alzheimer＇s disease （AD）, and its correlation with dementia severity. Methods The levels of MIF protein and mRNA in PBMC from 64 AD patients and 50 healthy subjects were measured using ELISA and Fluorogenic quantitative PCR, respectively. AD patients were classified into 3 groups according to MMSE scores as follows： mild, moderate and severe group. Results The expression levels of MIF were obviously higher in the AD group than in the control （ P 〈 0. 05 ） ; The expression levels of MIF were higher in the severe group than in the moderate or mild group （P 〈 0. 05 ） ; however, there were no significant difference in MIF expression levels between the moderate and mild group （ P 〉 0. 05 ）. A negative correlation between MIF expression levels and MMSE score was found in the AD patients（ r protein = - 0. 34, r nucleate = - 0. 36, P 〈 0. 01 ）. Conclusions MIF may play a critical role in pathological process of AD and its level correlates with the severity of dementia.
