摘  要:目的探讨雌激素受体β(estrogen receptorβ,ERβ)在大鼠结肠的表达及其对结肠肌电、Cajal间质细胞(interstitialcells of Cajal,ICC)的影响。方法免疫组化观察ERβ在正常大鼠结肠的表达和分布。20只大鼠随机分为两组,实验组给予他莫昔芬(tamoxifen,TAM)灌胃,3个月后测定结肠慢波频率及振幅;免疫荧光染色观察肠壁ICC数目的变化。结果ERβ在大鼠结肠黏膜、黏膜下及肌间神经丛表达。TAM拮抗雌激素后,大鼠结肠慢波频率及振幅均明显降低(P<0.05),肌间神经丛及黏膜下神经丛ICC数目较对照组明显减少(P<0.01)。结论ERβ信号通路可能通过影响ICC,从而导致结肠动力减弱。Objective To investigate the expression of ERβ and its effect on colonic motility and interstitial ceils of Cajal in rats. Methods Immunohistochemistry methord was used to detect the distribution and expression of ERβ in rats. Frequency and amplitude of colonic slow waves were examined after the animals fed for 3 months. The number of ICC were observed by fluorescence staining. Results ERβ was detected in mucous layer, myenteric nerve plexus and submucous nerve plexus in colon. In comparison with the control group,the frequency and amplitude of slow waves of experimental group were reduced significantly (P〈0.05) ,and the number of ICC also decreased (P〈0.01). Conclusion ERβ signaling pathway can influence the number of ICC and furthermore lead to STC.
