摘  要:目的:观察急性酒精中毒合并中度创伤性脑损伤后大鼠海马星形胶质细胞标记物胶质纤维酸性蛋白(GFAP)表达的变化。方法:健康成年雄性SD大鼠72只,随即机分为4组:假手术组(N组)、急性酒精中毒组(E组)、中度创伤性脑损伤组(T组)和急性酒精中毒合并中度创伤性脑损伤组(E+T组)。腹腔注射酒精(2.5g/kg)致使大鼠急性酒精中毒,2h后,按改进的Feeney's自由落体硬膜外撞击方法使其合并中度创伤性脑损伤(600g.cm)。各组动物术后6h、24h和48h处死。中性红染色观察海马CA1区神经元形态学改变;用免疫组织化学的方法检测海马CA1区GFAP表达变化。结果:与N组和E组相比,T组和E+T组GFAP表达显著增多(P<0.01)。术后6h和24h,T组GFAP表达显著高于E+T组(P<0.05);T组和E+T组的海马CA1区神经元细胞出现胞体肿胀,排列散乱,但T组上述形态学改变较E+T组明显。结论:急性酒精中毒合并中度创伤性脑损伤的早期可通过减少GFAP的表达,抑制星形胶质细胞激活,减少炎症反应发挥保护作用。Objective: To investigate the expression of GFAP in rat hippocampus after moderate traumatic brain injury following acute alcoholism. Methods: 72 healthy adult SD male rats were randomly divided into 4 groups: Group N (control group with sham-operated ), Group E (acute alcoholism ), Group T (moderate traumatic brain injury ) and Group E+T (moderate traumatic brain injury following acute alcoholism). Acute alcoholism was performed by intrapritoneal injection of alcohol (2.5g.kg^-1) in rats. Moderate traumatic brain injury was produced with the improved Feeney's method (600g.cm) at 2h after acute alcoholism. Animals were killed at 6h, 24h and 48h after trauma, and their brains were used for determination of the expression of GFAP by immunohistochemistry method and observation on morphological changes by toluylene red staining in CA1 area of hippocampus. Results: The expression of GFAP in Group T and Group E+T were higher than that in Group N and Group E (P〈0.01 ). The expression of GFAP in Group T were higher than that in Group E+T at 6 hours and 24hours (P〈0.05). By toluylene red staining, the neurons of Group T and Group E+T in CA1 area of hippocampus appeared body swelling and disorderly arrangement at 6 hours, and such morphologic change in group T appeared obvious compared with group E+T. Conclusion Acute alcoholism plays protective roles at early stage of moderate traumatic brain injury, which may be related to astrocyte inhibited by alcohol so as to reduce inflammatory reaction.
