摘 要:目的:观察氧诱导小鼠视网膜的病变情况。方法:出生7d的129Sv/C57BL6J小鼠随机分为实验组和对照组,各28只。实验组置(75±2)%高氧环境、室温(23±2)℃,日光照明,5d后返回空气环境;对照组置(23±2)℃空气环境中饲养。出生后第17d在视网膜平铺片及冰切片上通过免疫荧光法定量检测视网膜血管的增生情况,比较两组星型胶质细胞、小胶质细胞的差异,并通过TUNEL法检测细胞凋亡的情况。结果:出生后第17d实验组视网膜出现无血管区域,新生血管计数高于对照组(P<0.01);星型胶质细胞增多、密集,与相邻的血管间的联系紧密,其小胶质细胞出现活化和增殖,突起少而短,并有胞体肿胀现象;实验组小鼠视网膜存在细胞凋亡。结论:(75±2)%高氧环境、室温(23±2)℃成功诱导小鼠视网膜的病变。Objective: To observe retinal pathological changes of oxygen-induced mice. Methods: A total of 56 one-week-old neonatal 129Sv/C57BL6J mice were randomly divided into experimental group( n=28 )and control group( n=28 ). The experimental group was exposed to 75%±2% oxygen environment and the room temperature controlled at 23℃ 20℃, giving daylight illumination. These mice were back to natural air environment after 5 days. The control group was fed only in natural air environment. On the seventh day after birth, the retinal vascular proliferation was examined quantitatively with immunofluorescence method in the retinal vascular digest preparation and frozen sections. The astrocytes and microglias were compared between the two groups. Apoptosis of retina cells were detected by TUNEL method. Results: On the seventh day after birth, there were non vascular regions and apoptosis in the retina of the mice in the experimental group, and neovascularization count of experimental group was higher than that in the control group. In the experimental group, astrocytes increased and was closely related to contiguous vessels and its microglias was activated with less and shorter processes and swelling of cell bodies. Conclusion: 75%± 2% oxygen environment and the room temperature controlled at 23± 20℃ can induce retinopathy.
