补脾益气中药对哮喘大鼠模型BALF中IL-5GM-CSF TNF-α和sIL-2R的影响

摘　　要：目的:探讨白细胞介素-5(IL-5)、粒细胞-巨噬细胞集落刺激因子(GM-CSF)、肿瘤坏死因子(TNF-α)和可溶性白细胞介素-2受体(sIL-2R)在哮喘发病过程中的作用及补脾益气中药(补脾益气中药)对其含量的影响。方法:采用卵蛋白等致敏大鼠哮喘模型,观察支气管肺泡灌洗液(BALF)中IL-5、GM-CSF、TNF-α和sIL-2R含量的变化。结果:与正常组比哮喘组BALF中IL-5、GM-CSF、TNF-α和sIL-2R含量均显著增高,尤其是IL-5增高更显著;相关分析表明GM-CSF和IL-5、GM-CSF和TNF-α显著相关;补脾益气中药能降低BAFL中这4种细胞因子的含量。结论:IL-5、GM-CSF、TNF-α和sIL-2R在哮喘大鼠的气道炎症细胞浸润过程中有很重要的作用,它们加速了哮喘非特异性气道炎症的发生和发展。GM-CSF与IL-5和TNF-α之间存在密切的联系,可能对嗜酸性粒细胞(EOS)的聚集、迁移、活化和黏附有协同作用。补脾益气中药可以通过降低细胞因子水平,改善气道的炎症状况。objective ：The action and their relationship of interleukin -5 （IL -5 ）, granulocyte - macrophage colony - stimulating factor （ GM - CSF）, tumor necrosis factor （ TN F - α） and soluble interleukin - 2 receptor（ sIL - 2R） during the course of asthma attack is discussed and the effect of their contents by Spleen - Qi Nourishing Chinese Herbal （ Add Ingredients Jade Screen Powder）. Methods ： A sensitized asthmatic rat model is obtained by employing egg albumin to it. The change of the content of IL -5, GM - CSF,TNF - α and sIL - α in broncho - alveolar lavage fluid（BALF） is observed. Results ： In contrast to the control group, the content of the asthma group in BALF has a dramatic growth, especially the content of IL - 5. Correlation analysis shows that GM - CSF and IL - 5, GM - CSF and TNF - are significantly correlated, respectively. Add Ingredients Jade Screen Powder can degrade the contents of four ＇cell factors in BALF. Conclusion ：IL -5 ,GM - CSF,TNF -α and sIL -2R are of great importance during the course of infiltration of the airway inflammation cell of the asthmatic rat. They accelerate the emergence and development of the nonspecific airway in- flammation of asthma. There exists close link between IL - 5, TNF - α and GM - CSF, and it is expected that this has synergistic action to the aggregation, migration, activation and adherence of the eosinophil （EOS）. Add Ingredients Jade Screen Powder can depress the level of cell factor to improve the airway＇s inflammation state.
