摘 要:目的观察网络抗氧化剂对大鼠非酒精性脂肪性肝炎的预防与治疗效果。方法雄性SD大鼠40只随机分为4组:正常对照组、模型组、网络抗氧化剂预防组、网络抗氧化剂治疗组(各10只),空白对照组喂以正常饲料,其余3组均喂以高脂饮食。网络抗氧化剂预防组在高脂饮食同时给予维生素E(100mg/kg)、硫辛酸(40mg/kg)、辅酶Q10(100mg/kg)、维生素C(100mg/kg)、还原型谷胱甘肽(100mg/kg)联合灌胃治疗,每日1次。网络抗氧化剂治疗组在高脂饮食12周(造模成功)后开始给予上述网络抗氧化剂联合灌胃治疗。16周时处死各组大鼠,观察肝功能和血脂变化,检测血清及肝组织丙二醛(MDA)和超氧化物歧化酶(SOD)活性。结果网络抗氧化剂预防组及治疗组体重、肝湿重、血清丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、MDA、甘油三酯(TG)、总胆固醇(TC)及肝组织MDA均显著低于模型组,血清及肝组织SOD活性显著高于模型组。网络抗氧化剂预防组体重、肝湿重、ALT、TG和血清MDA与网络抗氧化剂治疗组比较差异无统计学意义,血清AST、TC和肝组织MDA均显著低于治疗组,血清及肝组织SOD活性显著高于治疗组。结论网络抗氧化剂对大鼠非酒精性脂肪性肝炎有较好的防治作用。Objective To investigate whether the network antioxidants has empodistic and therapeutic effects on nonalcoholic steatohepatitis. Methods 40 male rats were randomly divided into control group (ordinary diet), model group (high-fat diet),network antioxidants empodistic group (high-fat diet plus intragastric administration with 40 mg/ kg lipoic acid, 100 mg/kg coenzyme Q10, 100 mg/kg vitamin C, 100 mg/kg vitamin E and 100 mg/kg glutathione daily) and network antioxidants therapeutic group(intragastric administration with above-mentioned antioxidants after feeding high-fat diet 12 weeks). Weight gain, liver index, serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), total cholesterol(TC), triglyeride(TG), serum and hepatic malondialdehyde (MDA) and superoxide dismutase (SOD) were examined at the end of 16 weeks. Results Body weight, liver humid weight, serum ALT, AST, TC, MDA and hepatic MDA of network antioxidants-empodistic and treated rats were significantly lower than those of model group, while serum and hepatic SOD activities were significantly higher than those of model group. Serum AST, TC and hepatic MDA of network antioxidants-empodistic rats were significantly lower than those of treated group. Serum and liver SOD activities of network antioxidants-empodistic rats were significantly higher than those of treated group but the body weight, liver humid weight, serum ALT, TG and MDA have no difference between these two groups. Conclusion Network antioxidants have an empodistic and therapeutic efficacy on the rats with nonalcoholic steatohepatitis.
