摘  要:目的:探讨葡萄糖调节蛋白78(GRP78)在高低转移对的乳腺癌细胞的表达及其意义。方法:分别采用半定量RT-PCR,蛋白免疫印迹法和细胞免疫荧光方法检测GRP78在三种不同乳腺癌细胞株(MDA-MB-231、435S和MCF-7)的mRNA蛋白表达及其亚细胞定位情况。结果:GRP78 mRNA和蛋白在(MDA-MB-231细胞的表达最高,其次为(MDA-MB-435S,在MCF-7的表达最低;共聚焦显微镜下观察到GRP78蛋白主要定位在细胞浆,在细胞膜以及细胞核内也有少量的表达。结论:GRP78是乳腺癌的一个促癌基因,在乳腺癌的发生、发展过程中发挥着重要作用,其可作为抗乳腺腺癌治疗的一个新的分子靶点。Objective: To evaluate the expression of glucose-regulated protein 78 (GRP78) in three breast cancer cell lines (MDA-MB-231, MDA-MB-435S, amt MCF-7) and to determine its clinical significance. Methods: Reverse transcriptase-polymerase chain reaction (RT-PCR) and Western blot were used to amplify GRP78 mRNA and detect protein expression. Using confocal microscopy, we observed the subcellular localization of GRP78. Results: RT-PCR revealect that the expression of GRP78 mRNA was higher in the MDA-MB-231 breast cancer cells than in MDA-MB- 435S and MCF-7 cells. The results of Western blot indicated that GRP78 protein expression showed a pattern similar to that of GRP78 mRNA expression across the three groups with the expression of GRP78 protein in MDA-MB-231 cells as the highest among the three groups. GRP78 protein was mainly expressed in the cytoplasm, membrane and nucleus. Limited expression of GRP78 protein was observed in MCF-7 cells. Conclusion: GRP78 may play a key role in the carcinogenesis and development of breast cancer.
