摘  要:目的:研究胶质瘤中蛋白激酶PKC-α mRNA表达与NDRG2 mRNA表达的相关性,寻找可能与NDRG2相互作用的上下游分子.方法:用实时定量PCR技术检测26例脑胶质瘤及相应瘤旁组织中NDRG2和PKC-α mRNA表达量.结果:26例脑胶质瘤NDRG2 mRNA表达低于瘤旁组织(0.95±0.16vs1.54±0.18,P<0.05),PKC-α mRNA表达高于瘤旁组织(1.23±0.11vs0.97±0.18,P<0.05),且随胶质瘤恶性程度增高,NDRG2表达逐渐减少(P<0.05),PKC-α表达有逐渐增加的趋势.NDRG2与PKC-α表达无相关(P>0.05).结论:NDRG2,PKC-α可能与胶质瘤的发生发展及恶性演变有关,在mRNA表达水平,还不能认为NDRG2与PKC-α之间有相关性.AIM: To investigate the correlation between PKC-α and NDRG2 expressions in human glioma, so as to find the upstream or downstream molecular interacting with NDRG2. METHODS: Real time PCR was used to analyze the expressions of PKC-α and NDRG2 mRNA in 26 pairs of glioma and adjacent normal tissues. RESULTS: Compared with adjacent normal tissues, the expression level of NDRG2 mRNA in glioma tissues was reduced significantly [(0. 95±0. 16) vs (1. 54±0. 18), P 〈 0.05, but PKC-α was increased significantly ( 1.23±0.11 ) vs (0.97±0. 18 ), P 〈 0.05 ]. And with pathologic grade increasing, there was a trend for NDRG2 level to reduce ( P 〈 0.05) , however PKC-α level to increase ( P 〉 0.05). But there was no statistical correlation between PKC-α and NDRG2. CONCLUSION : The aberrant expressions of NDRG2 and PKC-α are possibly associated with the malignant transformation of glioma, but at mRNA level, PKC-α possibly has no correlation with NDRG2 in glioma.
