摘 要:目的:探讨不同期别乳腺癌组织中DNA依赖蛋白激酶(DNA-PK)的催化亚单位DNA-PKcs的表达变化情况。方法:采用免疫组化SP法检测Ⅰ~Ⅳ期乳腺癌组织各30例中的DNA-PKcs表达情况。结果:DNA-PKcs在Ⅰ期乳腺癌组织中呈高表达,在Ⅱ期中的表达有降低趋势(与Ⅰ期比较P<0.01,与Ⅲ期比较P>0.05,与Ⅳ期比较P<0.01),在Ⅲ、Ⅳ期乳腺癌组织中的表达明显低于Ⅰ期乳腺癌,P均<0.01。结论:DNA-PKcs在不同阶段乳腺癌中的表达情况是随着乳腺癌的分期变化而变化,从早期到晚期呈逐渐降低趋势,可能与乳腺癌的分期及预后有一定相关性。OBJECTIVE: To investigate the interrelation between the expression changes of DNA-PKcs and stages in breast cancer. METHODS: The expression of DNA-PKcs was examined in 30 cases of stage Ⅰ breast cancer tissues, 30 cases of stage Ⅱ breast cancer tissues, 30 cases of stage Ⅲ breast cancer tissues and 30 cases of stage Ⅳ breast cancer tissues by using the immunohistochemical SP method. RESULTS: The expression levels of DNA-PKcs in stage Ⅰ breast cancer tissues were high. Compared with stage Ⅰ breast cancer, the expression of DNA-PKcs in stage Ⅱ breast cancer was low, P〈0.01. Simultaneously, the expression of DNA-PKcs between stage Ⅱ and stage Ⅳ had a significant difference, P〈0.01. The expression levels of DNA-PKcs in stage Ⅲ and stage Ⅳ breast cancer tissues were significanfly lower than those in stage Ⅰ breast cancer tissues, both P〈0.01. CONCLUSIONS:From early breast cancer to advanced breast cancer, there is a tendency that the expression of DNA-PKcs in breast cancer gradually decreases. The low-expression of DNA-PKcs may play a role in the development and metastases of breast cancer. DNA-PKcs could be associated with breast cancer staging system and prognosis.
