摘  要:目的:探讨玻璃体腔内注射外源性睫状神经营养因子(CNTF)是否对大鼠视神经中度不全损伤视网膜神经节细胞(RGCs)具有保护作用。方法:采用无创血管钳对成年大鼠造成视神经中度不全损伤,实验组伤后即时1、2、4周分别向玻璃体腔内注射CNTF溶液2μl;同实验组操作,对照组在每个时间点向玻璃体腔内注射2μl双蒸水。在伤后1、2、4、8周时分别做生长相关蛋白-43(growth associated protein-43,GAP-43)免疫组织化学检测和光镜的组织学观察。结果:①伤后1周对照组和实验组视网膜均有水肿,RGCs肿胀。伤后2周2组视网膜神经节细胞层(GCL)均变稀疏,实验组水肿明显减轻。伤后4周水肿消退,GCL层细胞数明显下降。8周实验组视网膜厚度正常,对照组各层变薄。②伤后1周对照组和实验组GAP-43在GCL层表达呈阳性。伤后2、4、8周GAP-43在GCL层表达至高峰。实验组明显强于对照组(P<0.01)。结论:CNTF对大鼠视神经损伤后RGCs具有明显的保护作用。Objective:To observe the protective effect of exogenous CNTF on the retinal ganglion cells following incomplete optic nerve injury in rats by intravitreous injeetion.Methods:The optic nerve was crushed by a well defined forceps to set up an animal model with incomplete moderate optic nerve injury in rats.CNTF.A moderate optic nerve injury in some rats was by intravitreal injections of 2 μg CNTF,and control groups were injected withdistilled water and used as control group at immediately followed time 1,2,4,8 week after the injury.Rats were sacrificed at the first,the 2,4,8 week after optic nerve iniury.The expression of GAP-43 in retina was determined by immunohistochemical techniques at the first 2,4,8 week after optic nerve iniury.Light microseope and transmission elcetronmieroseope examination was taken for the microstrueture of retina.Results:①The expression of GAP-43 in retina following moderate optic nerve injury in CNTF treating groups was higher than that in the control group at the 2,4,8 week after optic nerve injury,the(P〈0.01).②After the first,2,4,8 week after optic nerve injury,the comparison between the RGCs number of treating groups and control groups is very prominent.Both of two groups of the RGCS'number reduced on different degree by light microseope to observe.③After the first,2,4,8 week after optic nerve injury,Under electron microscopy the RGCs conformation of CNTF treating groups is radical intergrity,it′s better than control groups.chromatin aggregation of RGCs became conspicuous.Cell bodies were prominently furrowed.Condensed nucleipresented in the retina.Cytoplasmic atrophy was profound,and nucleus membrane was in its integrity.Other injured ganglion cells membranes might be dissolved and showed dropsy and disintegration of organelle.Conclusion:The intravitreal application of rhCNTF can enhance the RGCs long-term survival and may play an important role in the regeneration of axons following a moderate incomplete optic nerve injury in adult rats.
