巯基乙醇在家蝇幼虫血淋巴中适宜添加量研究

摘  要:[目的]为家蝇幼虫血淋巴抗菌活性物质的分离、纯化提供依据。[方法]以大肠杆菌为指示菌,利用琼脂糖孔穴扩散法研究巯基乙醇对家蝇幼虫血淋巴抑菌活性的影响,确定巯基乙醇作为抗氧化剂在家蝇幼虫血淋巴中的适宜添加量。[结果]巯基乙醇能抑制大肠杆菌的生长,抑菌效果随其浓度的增加更为明显。当巯基乙醇体积浓度≤10-6时,其对大肠杆菌无抑菌效果,说明巯基乙醇的最大非抑菌体积浓度为10-6。体积浓度为10-6的巯基乙醇可有效保护浓度小于或等于1∶5的家蝇幼虫血淋巴悬液不被氧化,且其对大肠杆菌的抑菌效果不受影响。[结论]巯基乙醇在家蝇幼虫血淋巴中的适宜加入量应根据试验中具体情况确定。[Objective] The research aimed to provide the basis for the isolation and purification of antimicrobial active substance in haemolymph of housefly larva.[Method] With Escherichia coli as indicative bacteria,the effects of mercaptoethanol on the antibacterial activity of haemolymph of housefly larva were studied by using agarose cavity diffusion method.And the optimum adding amount of haemolymph as antioxidant in haemolymph of housefly larva was confirmed.[Result] Haemolymph could inhibit the growth of E.coli and the antibacterial effect was more obvious with the increasing of its concn.When the concn.of haemolymph was less than or equal to 10-6 ml/ml,it had no antibacterial effect on E.coli,which indicated that the maximum un-antibacterial concn.of haemolymph was 10-6 ml/ml.Haemolymph at 10-6ml/ml could protect the haemolymph suspension of housefly larva at the concn.of less than or equal to 1:5 from being oxidated and its antibacterial effect on E.coli was not affected.[Conclusion] The optimum adding amou nt of haemolymph in haemolymph of housefly larva should be confirmed according to the concrete conditions in the test.
