经肝动脉化疗栓塞术对肝癌细胞凋亡抑制基因Survivin mRNA和蛋白的影响

摘　　要：目的探讨肝动脉化疗栓塞术(TACE)对肝细胞癌(HCC)细胞凋亡抑制基因Survivin mRNA和Survivin蛋白的影响。方法HCC患者分组:术前TACE组(A组,26例),单纯手术组(B组,30例),正常肝组织组(C组,16例)。应用原位杂交法、抗生物素蛋白链菌素-过氧化物酶(streptavidin peroxidase,S-P)免疫组织化学法、脱氧核糖核苷酸末端转移酶介导的缺口末端标记(terminal deoxynucleotidyl transferase mediated dUTP nick end labeling,TUNEL)法进行分析。结果(1)A组癌组织Sur-vivin mRNA和Survivin蛋白阳性表达率、灰度差值<B组(P<0.05);A、B组癌旁组织、C组均无Survivin mRNA和蛋白表达。(2)A组癌组织的凋亡指数高于B组癌组织和C组(P<0.05);B组癌组织的凋亡指数高于C组(P<0.05)。结论TACE治疗可使HCC中Survivin mRNA和蛋白表达减少、凋亡指数增加。Objective To study the effect of transcatheter arterial chemoembolization（TACE） on inhibitor apoptosis protein - survivin and survivin mRNA in hepatocellular carcinoma （ HCC ）. Methods Patients with HCC were divided into preoperative TACE group （ group A, 26 cases ） ,only operation group（ group B ,30 cases ） and normal liver tissue group（ group C, 16 cases ）. The situ hybridization, immunohistochemical S- P method and terminal deoxynucleotidyl transferase mediated dUTP nick and labeling（ TUNEL） were used in this study. Results （ 1 ） The positive expression and the grey difference of survivin mRNA and survivin in the tissues of carcinoma of group A were more than that in group B （P 〈0.05 ）. The was no expression of survivin mRNA and survivin in the matched adjacent nontumor tissues in group A and B and the normal liver tissues. （ 2 ） The apoptosis index （ AI ） in the tissues of carcinoma of group A was more than the tissues of carcinoma in group B and C（P〈0.05）.The AI in the tissues of carcinoma in group B was more than that in group C （P〈0.05）.Conclusion TACE can decrease the expression of survivin mRNA and survivin,and can increase the apoptosis index.
