摘  要:目的探讨垂体瘤转化基因(PTTG)和血管内皮生长因子(VEGF)在垂体瘤中的表达及其与肿瘤血管形成的关系,为进一步研究垂体瘤的抗血管治疗提供理论依据。方法利用免疫组化方法检测48例垂体瘤组织中PTTG、VEGF及微血管密度(MVD)的表达,并分析PTTG、VEGF的相关性以及与MVD和垂体腺瘤侵袭性的关系。结果PTTG和VEGF在垂体腺瘤中的阳性表达率分别为66.7%(32/48)和79.2%(38/48),PTTG和/或VEGF阳性表达的垂体腺瘤组织中的MVD值均显著高于PTTG和VEGF阴性者(P<0.05),PTTG的表达与VEGF的表达呈明显正相关(P<0.05);PTTG、VEGF的表达与垂体腺瘤的侵袭性有关(P<0.05)。结论PTTG、VEGF对肿瘤血管形成起重要作用,可能通过PTTG-VEGF调节旁路途径促进垂体腺瘤的肿瘤血管形成;PTTG、VEGF的表达可作为判断垂体腺瘤侵袭性的一项生物学指标,对判断预后也有一定的意义。Objective To investigate the expressions of pituitary tumor transforming gene (PTrG) and vascular endothelial growth factor (VEGF) and their roles in the regulation of angiogenesis in human pituitary adenomas in order to provide the theoretical basis for treatment of invasive pituitary adenomas by anti-angiogenesis. Methods Expressions of PTrG, VEGF and microvessel density (MVD) were determined by immunohistochemical technique in 48 cases of human pituitary adenomas. The relationship between the expressions of PTrG and VEGF and the relationship of invasiveness of pituitary adenomas with expressions of PTrG and VEGF were analyzed. Results The positive rates of PTrG and VEGF expressions were 66.7% (32/48) and 79.2% (38/48) respectively. The expression of MVD was significantly higher in the PTrG-positive or VEGF-positive groups than that in the negative ones (P〈0.05). The expression of PTrG was positively correlated with the expression of VEGF (P〈0.05). The expressions of PTrG and VEGF were positively correlated with invasiveness of pituitary adenoma (P〈0.05). Conclusions It is suggested that the expressions of PTrG and VEGF play important roles in the angiogenesis of pituitary adenomas. They may serve as an assistant indicator of the invasiveness and are helpful to judging the prognosis in the patients with pituitary adenomas.
