摘 要:目的探讨同种异体NK细胞对CD3+4早期急性髓系白血病(AML)细胞的体外杀伤活性。方法免疫磁珠法分离5例健康个体NK细胞,以NK杀伤敏感细胞株K562作为对照,乳酸脱氢酶(LDH)释放法测定不同效靶比时NK细胞对CD3+4早期AML细胞KG1a的杀伤活性。结果AML细胞株KG1a中CD34抗原表达率为98.0%±1.1%,分选后的NK细胞纯度为93.2%±3.7%。不同效靶比时NK细胞对KG1a细胞均有杀伤活性,且随着效靶比的增高,其杀伤活性增高(P<0.05)。结论同种异体NK细胞对CD34+早期AML细胞具有一定的杀伤活性。Objective To study cytotoxicity in vitro of aUogenetic NK cells on human CD34 early acute myelogenous leukemia cells. Methods Expression of CD34 antigen on acute myelogenous leukemia cell line-KGla cells was detected by FACS. Cytolysis against KGla cells of NK cells isolated from 5 healthy persons with MACS was detected by LDH releasing assay at different effect-to-target ( E: T) ratios, compared with K562 which was sensitive to NK cells. Results Expression of CD34 antigen on KGla cells was 98.0% ± 1.1% and purity of NK cells after isolated was 93.2% ±3.7% detected by FACS. There was cytotoxiclty in vitro of aUogenetic NK cells against KGla cells at different E: T ratios. With increasing of E: T ratios, cytotoxicity of allogenetic NK cells against KGla cells was increased (P 〈 0. 05). Conclusion Allogenetic NK cells display cytotoxicity against CD34^+ early acute myelogenons leukemia cells.
