摘 要:目的探讨靶向survivin基因RNAi对腺样囊性癌增殖的抑制效应。方法设计、合成siRNA(small interfering RNA),构建表达载体pGenesil-shRNA-survivin,将重组质粒导入人腺样囊性癌ACC-M细胞株。采用逆转录-聚合酶链反应(RT-PCR)、Western-blot法分别检测转染后的ACC-M细胞中survivin mRNA和蛋白表达的变化,用噻唑蓝(MTT)法来检测细胞的增殖。结果测序证实表达载体构建成功,在pGenesil-shRNA-survivin的ACC-M细胞株中,survivin mRNA及蛋白表达明显降低,ACC-M细胞增殖受到抑制。结论pGenesil-shRNA-survivin重组质粒能有效抑制腺样囊性癌细胞增殖。Objective To investigate inhibitory effect of siRNA on survivin in Adenoid cystic carcinoma cells lines. Methods We designed and synthesized siRNA . Eukaryotic expression vector pGenesil-shRNA- survivin was constructed . Recombinant Plasmid was transfected into ACC-M cells. The expression of survivin mRNA and protein was detected by RT-PCR and Western-blot. The propagation of Adenoid cystic carcinoma cells was detected by MTT. Results The recombinant plasmid was successfully constructed . The expression of survivin mRNA and protein was obviously reduced . ACC-M cells propagation was inhibited. Conclusion Recombinant plasmid pGenesil-shRNA-survivin can specially suppress adenoid cystic carcinoma cells propagation.
