摘 要:目的研究高氧保存液对大鼠供肝的保护作用。方法采用大鼠肝脏非循环离体灌注模型,分别以乳酸钠林格氏液和高氧乳酸钠林格氏液对大鼠肝脏低温保存0、1、36、h,随后进行常温灌注30 min,测定灌注流出液氧自由基代谢产物丙二醛(MDA)和超氧化物歧化酶(SOD)的含量、肝细胞内Ca2+浓度;观察肝脏组织形态学变化。结果在同一时段,高氧乳酸钠林格氏液组大鼠肝脏细胞内Ca2+浓度及流出液MDA含量较乳酸钠林格氏组低,而SOD含量较高。光镜、电镜观察两组肝脏组织形态学变化也有一定的差别。结论高氧乳酸钠林格氏液对大鼠供肝保护作用优于乳酸钠林格氏液,即高氧保存液对供肝有更好的保存作用。Objective To study the protective effect of hyperoxic preservative fluid on donor liver. Methods Using the non-circulated isolated perfused rat liver (IPRL) model, rat livers were preserved in hypothermy for 0,1,3 and 6 h with natrium lacticum ringer's solution or hyperoxic natrium lacticum ringer's solution, then perfused in common temperature 30 min. The effects were assessed by measuring the content of malonaldehyde and superoxide dismatase in the efflu- ent and content of intracellular calcium ion in the rat hepatocytes, observing the morphological change in the liver. Results The content of intracellular calcium ion and MDA in the rat hepatocytes preserved by hyperoxic natrium lacticum ringer's solution was less, however the content of SOD was higher, than that in those preserved by natrium lacticum ringer's solution. Morphological changes were different between the two groups. Conclusion Protective effect of donor liver preserved by hyperoxic natriun lacticum ringer's solution may be better than preserved by natrium lacticum ringer's solution.
