摘 要:目的通过动物实验研究云芝多糖(CVP)对实验性肝损伤的防治机制。方法昆明种小鼠分为对照组、模型组和不同浓度的CVP保护组,饲养7d后对照组腹腔注射调和油溶液,其余各组腹腔注射0.15%CCl4调和油溶液(10mL/kg体重),24h后眼球取血分离血清,用赖氏法测ALT和AST。制备10%的肝匀浆测MDA、GSH、SOD、NOS和NO的含量。结果CCl4肝损伤时,不同浓度的CVP可降低血清中ALT和AST的活性(P<0.05),增加SOD的活性和GSH的含量(P<0.01),降低NOS的活性和MDA、NO的含量(P<0.01),体外抗氧化实验CVP可抑制脂质过氧化产物MDA的生成。结论CVP能有效的清除自由基,诱导SOD和GSH的合成,降低NOS活性和NO的含量,对实验性肝损伤有保护作用。[Objective] To find out the protectve effect of Coriolus versicolor polysaccharides on liver injury through animal experiment. [Methods] Kunming mice were divided into three groups: control group, model group, group treated with CVP. After being fed for 7 days, mixed-edible-oil solution was injected into group control through belly cavity, group model and group treated with CVP were injected 0.15% CCl4 mixed with mixed-edible-oil. 24 hours after injection, serum from mice's eyeballs was examined for ALT and AST levels by using Reitman method. 10% live preparation was tested for the levels of MDA, GSH, SOD, NOS and NO. [Results] Different levels of CVP decreased the levels of ALT and AST (P〈0.05), increased the levels of serum SOD and GSH (P〈0.01)and decreased the levels of MDA and NO in liver-damaged mice by CCl4. [Conclusion] CVP can eliminate free radicals, induce the synthesis of SOD and GSH, decrease NOS activity and NO content, and protect mice from liver damage.
