摘  要:目的研究PTEN/MMAC1/TEP1、TGF-β1在前列腺癌及前列腺增生中的表达探讨前列腺癌组织中PTEN/MMAC1/TEP1、TGF-β的表达与临床病理特征和生物学行为的关系及其意义。方法前列腺癌标本26例,前列腺增生标本18例,均为存档石蜡切片。应用原位杂交技术研究PTEN/MMAC1/TEP1表达水平,应用免疫组化研究PTEN/MMAC1/TEP1、TGF-β1表达。结果用原位杂交方法:前列腺癌组织中PTEN/MMAC1/TEP1表达42.31%,前列腺增生组织中的表达94.44%(P<0.01)。而免疫组化方法前列腺癌组织表达为34.62%,前列腺增生为94.44%(P<0.01)。组织学分级为Ⅰ、Ⅱ级的表达为72.72%高于Ⅲ级的20.00%(P<0.05)。临床分期A-C期的表达为66.67%,D期为21.43%(P<0.05)。表达为阳性的血清PSA浓度均值为(29.36±1.74)高于阴性的(15.92±2.06)(P<0.01);TGF-β1表达在前列腺癌组织为50.39%,在前列腺增生组织为11.11%(P<0.01)。组织学分级为Ⅰ、Ⅱ级的表达为27.27%低于Ⅲ级的73.33%(P<0.05)。临床分期A-C期的表达为25.00%,D期为78.57%(P<0.05)。表达为阳性的血清PSA浓度均值为(27.71±2.39),阴性的为(22.65±3.15)(P>0.05);前列腺癌组织中PTEN/MMAC1/TEP1、TGF-β1表达呈负相关(P<0.01)。结论用免疫杂交和原位杂交方法:前列腺癌组织中PTEN/MMAC1/TEP1表达低于前列腺增生组织中的表达(P<0.01)。表达与组织学类型、临床分期及血清PSA浓度有关(P<0.05);TGF-β1表达在前列腺癌组织高于前列腺增生组织(P<0.01)。TGF-β1表达与组织学类型及临床分期有关(P<0.05),而与血清PSA浓度无关(P>0.05);前列腺癌组织中PTEN/MMAC1/TEP1、TGF-β1表达呈负相关(P<0.01);PTEN的低表达可能增强了前列腺癌的侵袭和转移。[Objective] To investigate the expression of PTEN/MMAC1/TEP1, TGF-β1 in benign prostatic hyperplasia and prostatic cancer. To investigate the correlation with the pathological characteristic and biological behaviours of prostatic cancer. [Methods] A total of 18 cases of benign prostatic hyperplasia and 26 cases of prostatic cancer Paraffin-embedded specimens were studied. The expression of PTEN/MMAC1/TEP1 was valued by in situ hybridization and immunohistochemistry,and the expression of TGF-β1 were investigated by immunohistochemistry. [Results] The positive rate of PTEN/MMAC1/TEP1 expression was 42.31% in Pca, 94.44% in BPH (P 〈0.01). In Ⅰ -Ⅱ histological degree was 72.72%, and 20% in Ⅲ degree (P 〈0.05).in A-C stage was 66.67% and D stags was 21.43% (P 〈0.05). The concentration of serum PSA was (29.36±1.74) in positive cases, and (15.92±2.06) in negative cases (P 〈0.01). The positive rate of TGF-β1 expression was 50.39% in Pca, 11.11% in BPH (P 〈0.01). In Ⅰ -Ⅱ histological degree was 27.27%, and 73.33% in Ⅲ degree (P 〈0.05). That of A-C stage was 25% and D stags Was 78.57% (P 〈0.05). The concentration of serum PSA was (22.71±2.39) in positive cases, and (22.65±3.15) in negative cases (P 〉0.05). [Conclusions] The expression of PTEN/MMAC1/TEP1 was found significantly different between Pca and BPH. There is association between the expression of PTEN/MMAC1/TEP1 and histological degree, clinical stage and the concentration of serum PSA in prostatic cancer. The expression of TGF-β1 is found significantly different between Pea and BPH.There is association between the expression o.f TGF-β1 and histological degree,clinical stage in prostatic cancer.There is no association with the concentration of serum PSA in Pea. There is negative correlation between expression of PTEN/MMAC1/TEP1 and TGF-β1. The lower expression of PTEN/MMAC1/TEP1 may enhance the metastasis of prostatic cancer.
