摘  要:目的观察脑缺血再灌注后大鼠胰岛素生长因子-1(IGF-1)及其受体(IGF-1R)表达对其神经行为的影响。方法成年健康雄性Wistar大鼠28只,应用线栓法经左侧颈外-内动脉插线建立大脑中动脉闭塞再灌注(MCAO/R)模型。应用BEDERSON评分法评价脑缺血再灌注后神经行为功能,免疫组织化学技术检测IGF-1和IGF-1R表达。结果脑缺血再灌注后各组动物均出现不同程度的神经功能受损,从再灌注3 d开始神经功能逐渐恢复,但至再灌注7 d才有显著性差异。免疫组化检测显示,脑缺血再灌注后脑组织IGF-1和IGF-1R表达区域和时相变化规律基本一致,再灌注12 h^7 d持续高表达,1~3 d达高峰。结论脑缺血再灌注后IGF-1和IGF-1R的早期表达可能对神经功能恢复有一定的促进作用。Objective To investigate the effects of insulin-like growth factor (IGF-1) and its receptor (IGF-1R) on neurobehavior after cerebral isehemie reperfusion in rats. Methods A middle cerebral artery oeelusion/reperfusion (MCAO/R) model was made in 28 healthy male adult Wistar rats, the neurobehavior of these rats was evaluated via Bederson,s score, and expressions of IGF-1 and IGF-1R were determined by immunohistoehemieal assay. Results All the MCAO/R animals appearred different extent of neurofunetional impairment. Recovery started from the 3rd day after reperfusion, its significant improvement on 7th day. The expressions of IGF-1 and IGF-1R lasted for 12 h-7 days after reperfusion, reached its peak for 1-3 days, which was ba sieally consistent in area and time-phase. Conclusion The early expressions of IGF-1 and IGF-1R might enhance the neurofunetional recovery after cerebral isehemie reperfusion.
