胃宁颗粒治疗胃癌前病变实验研究

摘　　要：目的探讨胃宁颗粒对胃癌前病变的治疗作用及可能机制。方法60只大鼠随机分成空白组、模型组、维甲酸组、胃宁颗粒低剂量组、胃宁颗粒中剂量组、胃宁颗粒高剂量组。观察治疗前后各组大鼠一般情况、胃黏膜病理组织学、端粒酶活性、细胞增殖指数(PI)、细胞凋亡指数(AI)的变化。结果胃宁颗粒各剂量组活动、饮食等一般情况好转,肠化生(IM)、不典型增生(ATP)、端粒酶阳性发生例数显著低于模型组(P<0.05或P<0.01);胃宁颗粒各剂量组细胞增殖指数(PI)与模型组相比显著降低(P<0.01),凋亡指数(AI)显著增高(P<0.05)。结论胃宁颗粒对胃癌前病变具有治疗作用,其机制可能与抑制端粒酶活性、促进癌前病变细胞凋亡抑制其增殖有关。Objective To study the effects and mechanisms of weining granule on precancerous lesions developed in gastric mucose of rate. Methods Sixty rats were randomly divided into six groups, including blank group, model group, retinoicacid group, high-dose weining granule group, middle-dose weining graunle group and low-dose weining granule group. General conditions, the pathological changes in stomach, telomerase activity,proliferation index（PI）, apoptosis index（AI） were measured. Results General conditions, such as activity and eating were improved in all weining granule groups. IM, ATP and telomerase activity were markedly decreased compared with model group. Incidences of PI and AI in all weining gronule group were markedly decreased than that in the control group （P 〈 0. 05 or P 〈 0. 01 ） ; Conclusion It was suggested that weining granule could treat precancerous lesions by suppressing telomerase activity and gastric epitheliums proliferation and accelerate cell apoptosis.
