肥大细胞类胰蛋白酶促进人乳腺癌细胞系MDA-MB-435的跨膜侵袭

摘　　要：目的研究类胰蛋白酶对人乳腺癌细胞系MDA-MB-435侵袭力的影响及其机制。方法通过体外细胞侵袭力试验观察不同浓度、不同时间类胰蛋白酶对乳腺癌MDA-MB-435细胞跨膜侵袭力的影响，并利用RT-PCR以及明胶酶谱法研究类胰蛋白酶对基质金属蛋白酶-2（matrix metalloproteinase-2，MMP-2）和金属蛋白酶组织抑制物（tissue inhibitor of metalloproteinases-2，TIMP-2）表达水平的调节。结果150～500pmol／L的类胰蛋白酶可以促进MDA-MB-435细胞的跨膜侵袭；还可以促进MMP-2、TIMP-2的mRNA表达（P〈0．01）和MMP-2的蛋白表达（P〈0．05）。结论类胰蛋白酶可以促进人乳腺癌细胞系MDA-MB-435细胞的跨膜侵袭，该作用可能与增加MMP-2的mRNA和蛋白表达、TIMP-2的mRNA表达相关。Objective To study the effect and mechanism of tryptase on the invasion of human breast carcinoma cell line MDA-MB-435.Methods We studied the effects of tryptase on the invasion of MDA-MB-435 cells in different concentrations and at different times.The effects of tryptase on the mRNA,protein expression of matrix metalloproteinase-2(MMP-2)and mRNA expression of tissue inhibitor of metalloproteinases-2(TIMP-2)were testified by RT-PCR and gelatin zymography.Results Tryptase(150 and 500 pmol/L)promoted the invasive activity of MDA-MB-435 cells,and upregulated the mRNA expression of MMP-2 and TIMP-2(P<0.01)and protein level of MMP-2(P<0.05).Conclusions Tryptase can promote the invasive activity of MDA-MB-435 cells,possibly mediated by upregulation of MMP-2 and TIMP-2 expression.
