摘 要:目的观察风疹病毒R16株体外感染人胚晶状体上皮细胞的生物学特性。方法利用相差显微镜、透射电子显微镜、免疫细胞化学技术观察风疹病毒R16株在人胚晶状体上皮细胞中的形态学及生长特性。结果风疹病毒R16株体外感染人胚晶状体上皮细胞后,细胞不产生明显的病变效应。受病毒感染的人胚晶状体上皮细胞生长速度较慢,10d后可在细胞质内观察到呈不规则球形、由单层脂质膜包绕、直径60~80nm的病毒颗粒,包括致密核心和无致密核心两种颗粒。而在胞核中未发现病毒颗粒。结论风疹病毒R16株能在人胚晶状体上皮细胞中增殖,为进一步研究风疹病毒感染后先天性白内障的发生机制打下基础。Objective To investigate the biological characteristics of the human embryonic lens epithelial cells infected by rubella virus R16 in vitro. Methods The phase contrast microscope, electron microscope and the techniques of irnmunocytochemistry were used to study the morphology and growth characters of the rubella virus R16 in the human embryonic lens epithelial cells. Results There were no obvious pathologic changes in the human embryonic lens epithelial cell changes after the rubella virus R16 infection. Those human embryonic lens epithelial cells infected by the virus grew slowly. The virus-related particles were observed in the cytoplasm near the nucleus at 10 d post infection. The virion was irregularly spherical, 60- 80 nm, with a mono-layered lipid membrane. The virions with or without dense core were usually seen around the nucleus. There were no virions or other virus-related particles in the nucleus. Conclusions Rubella virus R16 can reproduce in the human embryonic lens epithelial cells, which is the groundwork for the further study on the pathogenesis of the congenital cataract infected by the rubella virus.
