摘 要:蚕丝经过高温高压水脱胶处理、高温高压碱性水脱胶处理和中性蛋白酶水解后,制成了6种不同分子量范围的丝胶肽及其水解物。利用家蚕5龄蚕血液进行了丝胶肽抑制黑色素生成试验,研究结果表明这6种丝胶肽都具有抑制黑色素生成的作用。高温高压水处理获得的丝胶肽抗氧化作用最强,高温高压弱碱性水处理获得的丝胶肽抗氧化作用略低,而中性蛋白酶水解的丝胶肽对黑色素形成的抑制率最低。当丝胶质量分数达到0.5%,前二类丝胶的抑制率可分别达到55%和50%,而第三类酶解丝胶对黑色素生成的抑制率为20% ̄25%,仅为前二种的一半还不到。这些实验结果说明,高温或高温弱碱性处理的丝胶蛋白更适合于化妆品、护肤品、保健品等领域的应用。When silk fiber derived from Bombyx mori was subjected to degumming treatments in water and subsequent degraded processing in slightly alkaline aqueous solution under high-temperature and high-pressure, and neutral protease treatment in buffer, six kinds of the water-soluble silk sericin peptides and their hydrolytes with different molecular mass were obtained. Antioxidation tests of sericin were carried out in the fresh blood of 5th larvae Bombyx mori. The experimental results showed that the six kinds of sericin are of inhibition to the formation of black pigment in the fresh blood of 5th larvae. The inhibition rate of the sericin protein derived from the hot treatment under high-temperature in water is the highest of all, and the three kinds of sericin derived from the hot treatment under high-temperature in slightly alkaline water are moderate and those sericin derived from protease hydrolysis are the lowest of all. When 0.5% of sericin concentration used, the inhibition rates of the two formers are 55% and 50%, respectively, and the last sericin proteins from protease hydrolysis are 20-25%.These results indicated that those sericin proteins derived from high temperature in water or in slightly alkaline water are preferable to application to those fields including cosmetic, skin-care product, healthy food and so on.
