口腔颌面部肉瘤中hTERTmRNA和Ki-67表达的相关性研究

摘　　要：目的探讨口腔颌面部内瘤中hTERTmRNA和Ki-67表达的相互关系。方法应用原位杂交和免疫组化方法检测37例口腔颌面部内瘤和14例良性间叶组织肿瘤中hTERTmRNA和Ki-67的表达。结果口腔内瘤中hTERT-mRNA的阳性表达率明显高于良性肿瘤（P〈0．05），并随内瘤分化程度的降低而呈升高趋势，其中低度恶性内瘤与高度恶性内瘤比较差异有统计学意义（P〈0．05）。hTERTmRNA表达阳性内瘤具有较高的细胞增殖活性（Ki-67LI），明显高于hTERTmRNA阴性表达内瘤（P〈0．05）。结论hTERT基因激活和细胞增殖在口腔间叶组织细胞实现永生化、发生恶性变的过程中可能起重要作用。Objective To investigate the expression of hTERTmRNA and Ki -67 and their relationship in oromaxillofaeial sarcoma. Methods In 51 histologically diagnosed specimens of 37 sarcoma and 14 benign mesenehymal tumor from oral and maxillofaeial region, expression of hTERTmRNA and Ki - 67 was determined by immunohistochemistry and in situ hybridization. Results The positive rate of hTERTmRNA expression was significantly increased in oromaxillofaeial sarcoma compared to benign tumor （ P 〈 0.05） , and tended to increase with decrease of differentiation of oromaxillofaeial sarcoma. There was a significant difference of hTERTmRNA expression between sarcoma of low grade and high grade （ P 〈 0.05 ）. The hTERT - positive oromaxillofaeial sarcoma showed higer proliferative activity （as assessed by Ki -67 proliferating index） than the hTERT - positive （ P 〈 0.05 ）. Conclusion hTERT activation coupling with cell proliferation may play an important role during the course of immortalization and cancerization of oral mesenchymal cells.
