摘 要:目的:优化麦冬粗多糖除蛋白工艺。方法:参照Sevage方法回流除蛋白,以多糖中的葡萄糖含量和蛋白收率为指标,采用L9(3^4)正交试验设计对于除蛋白过程中三氯甲烷与正丁醇的体积比、回流时间、回流次数做了考察,确定麦冬粗多糖除蛋白的最佳工艺条件。结果:三氯甲烷与正丁醇的体积比和回流次数均为影响除蛋白的显著性因素,最佳工艺条件为:三氯甲烷与正丁醇的体积比6:1,回流时间为20min,回流次数为2次。结论:优化工艺稳定,且可有效提高多糖的纯度。Objective:To optimize the technology of removing protein from crude polysaccharide of Dwarf Lilyturf Root Tuber. Methods:Taking the Sevage technique for reference and taking the content of glucose in polysaccharide and the yield of protein as the observation indice to remove the protein by reflux. Using the L9 (34) orthogonal design, the ratio of chloroform/n-butyl alcohol, the length of time of reflux and the time of reflux were studied and the significant technical factors have been comfirmed. Results:The ratio of chloroform/n-butyl alcohol and the time of reflux are the significant factors. The optimizing parameters are. 6:1 (chloroform/n-butyl alcohol), 2 times,20 min of each time. Conclusion :This optimum technology is stable and efficient.
