摘  要:目的:检测转化生长因子β(TGF-β)/Smad信号转导途径中Smad2、Smad3和Smad4 mRNA在基底细胞癌(BCC)和鳞状细胞癌(SCC)中的表达水平。方法:应用反转录-实时定量聚合酶链式反应(PCR)方法分别检测BCC、SCC与正常对照皮肤中Smad2、Smad3和Smad4 mRNA的表达情况。结果:BCC和SCC皮损中Smad2、Smad3和Smad4 mRNA的表达水平均显著低于正常人对照皮肤。结论:皮肤BCC和SCC中Smad2、Smad3和Smad4表达下调可直接或间接导致TGF-β抑制上皮细胞异常增殖和转化的作用受阻,从而有助于上述表皮肿瘤的形成和发展。Objective: To investigate the expression of Smad2, Smad3 and Smad4 mRNA in basal cell carcinoma and squamous cell carcinoma. Melhods: The RT and quantitative real-time PCR were used to detect the expression levels of Smad2, Smad3 and Smad4 mRNA in basal cell carcinoma and squamous cell carcinoma specimem, and normal control skin as well. Resnlt: The expressiom of Smad2, Smad3 and Smad4 mRNA were significantly down-regulated in basal cell carcinoma and squamous cell carcinoma in comparison with those in normal control skin. Conclusion: Down- regulation of Smad2, Smad3 and Smad4 may interfere in TGF- β signaling transduction pathway directly or indirectly and contributes to the formation and progression of basal cell carcinoma and squamous cell carcinoma.
