摘  要:目的研究卒中易感型自发性高血压大鼠(SHR-SPs)脑组织中基质金属蛋白酶(MMPs)以及组织型基质金属蛋白酶抑制剂(TIMPs)表达与活性的变化。方法观察期间每两周测定各组大鼠的收缩压。6个月后,采用Kaplan-Meier生存分析比较两组大鼠的发病率和病死率;以蛋白质印迹及明胶酶谱法分析两组中存活大鼠脑组织MMP-2和MMP-9蛋白水平和活性,并采用蛋白质印迹及逆向明胶酶谱法分析存活大鼠脑组织TIMP-1和TIMP-2蛋白表达和活性。结果SHR-SPs组收缩压明显高于对照组(P<0.01);观察期间SHR-SPs组80%大鼠发生脑卒中,50%出现卒中性死亡;SHR-SPs中存活大鼠脑组织MMP-2蛋白表达及活性较对照组增高(P<0.05);存活SHR-SPs脑组织TIMP-2蛋白表达和活性明显高于对照组(P<0.05),TIMP-1无明显变化。结论SHR-SPs脑组织中MMP-2的表达及活性均增高,同时TIMP-2的含量及活性呈现代偿性增高。Objective To investigate expression and activity of cerebral matrix metalloproteinases and tissue inhibitors of matrix metalloproteinases in stroke-prone spontaneously hypertensive rats (SHR-SPs). Methods Systolic blood pressure (SBP) was measured in every two weeks. Incidences of stroke and mortality rate were compared by Kaplan-Meier survival analysis 6 months later. Western bolt, gelatin-zymography, and reverse-zymography were performed to detect protein expression, activity of gelatinases and their inhibitors in brain homogenates, respectively. Results SBP was greatly elevated in SHR-SPs at the end of follow-up(P 〈0.01 ). 80% of SHR-SPs were presented with stroke symptoms and mortality rate was 50%. The expression and activity of MMP-2 were increased compared with those of controls. The expression and activity of TIMP-2 were increased compared with those of controls, and TIMP-1 remained unchanged in all groups. Conclusion The results of present work showes that both the expression and the activity of MMP-2 in the brain of SHR-SP were significantly higher than control, and the expression and the activity of TIMP-2 are reciprocally elevated.
