摘  要:目的研究神经生长因子(NGF)的脑保护时间窗与半胱天冬酶-3(Caspase-3)表达的相关性。方法采用兔局灶性脑缺血再灌注损伤模型,分别于再灌注后Oh、1h、3h和6h将NGF立体定向导入梗死灶周,再灌注72h观察神经功能、梗死体积、灶周凋亡率和Caspase-3表达。结果再灌注0h、1h和3h灶周给予NGF,梗死体积分别较对照组下降50.1%、42.5%和35.2%,相应的灶周凋亡率及Caspase-3表达明显下降,神经功能恢复较好,用药越早越明显;再灌注6h给药,则无明显作用。相关分析显示梗死体积变化与Caspase-3表达具有明显相关性(P〈0.05)。结论NGF脑保护治疗时间窗与Caspase-3表达相关,抑制Caspase-3表达可能是NGF介导神经保护作用的机制之一。Objective To study the relationship between the therapeutic window of opportunity for NGF following focal cerebral ischemia-reperfusion injury in rabbits and the expression of cysteinyl aspirate- specific proteinase-3 (Caspase-3) protein. Method Focal cerebral ischemia-reperfusion model in rabbit was induced. NGF was administered at 0, 1, 3 and 6h after reperfusion respectively. Infarct volume, neurological deficit score, apoptotic percentage, and expression of Caspase-3 were assessed at 72h after reperfusion. Results NGF reduced infarct volumes compared with saline control group by 50. 1%, 42.5% and 35.2% when administered NGF at 0, 1 and 3h after reperfusion. The percentage of apoptotic cell and expression of Caspase-3 in the perifocal regions were significantly lower than that in control group when administered NGF within 3h after reperfusion (P 〈 0.01). There was no difference when administered NGF at 6h after reperfusion ( P 〈 0. 05 ). Infarct volumes were correlated significantly with expression of Caspase- 3 in the perifocal regions ( P 〈 0.01 ). Conclusions The effect of NGF on the expression of Caspase-3 in perifocal regions is related with the therapeutic time window. NGF-induced neuroprotective effect may be involved in reducing the expression of Caspase-3 protein.
