含睾丸酮的乳酸-乙醇酸共聚物微球的制备及体外药物释放

摘　　要：目的：对国内开发睾丸酮微球长效注射剂进行初步可行性研究。方法：以生物降解医用材料乳酸（ＬＡ）乙醇酸（ＧＡ）共聚物（ＰＬＧＡ，ＬＡＧＡ８∶２）为载体，用溶剂挥发溶剂萃取法制备含睾丸酮的ＰＬＧＡ微球，并通过体外药物释放试验评价睾丸酮ＰＬＧＡ微球的药物缓释作用。结果：采用溶剂挥发溶剂萃取法可得到收率、粒径大小和分布及含药量均满意的睾丸酮ＰＬＧＡ微球。结论：睾丸酮ＰＬＧＡ微球有望成为新的睾丸酮长效制剂。OBJECTIVE: To develop a biodegradable testosterone microspheres formulation. METHOD: Poly (D, L lactide co glycolide) (PLGA) microspheres containing testosterone was prepared by a solvent evaporation and solvent extraction process and the drug release test of the microspheres was carried our in vitro. RESULTS: The microspheres with good yield, disired size and satisfied drug loading was obtained, and a significant testosterone sustained release was shown in the test of drug release in vitro. CONCLUSION: PLGA microspheres containing testosterone can be prepared by a solvent evaporation and solvent extraction process. The microspheres is promising to be developed as a new long acting testosterone formulation.
