摘  要:以银杏花粉为试验材料,研究了花粉在离体培养条件下,培养基中蔗糖、硼、钙对其萌发率的影响,优化了培养基配方;并比较了3种不同的染色法对银杏花粉生活力的测定效果。结果表明:培养5d是进行花粉萌发率测定的最佳时期,蔗糖对银杏花粉萌发至关重要,在培养基中添加3%蔗糖可有效提高花粉萌发率,达到50%左右;硼和钙对银杏花粉萌发有一定的促进作用,最佳培养基配方为蔗糖3%+硼60mg/L+钙100mg/L。TTC染色法测定的银杏花粉生活力与离体萌发法测定的结果可以相互代替和印证,是一种较为准确的测定银杏花粉生活力的染色法。4℃冰箱干燥保存利于保持银杏花粉生活力。In order to test the pollen vitality of Ginkgo biloba, different isolated culture medium with added Ca^2+, boric acid and sucrose were used. And three different staining tests were used to determine pollen vitality of Ginkgo biloba. Meanwhile differences of the methods on pollen vitality identification were studied. We also studied the relationship between storage methods and vitality of Ginkgo pollen. The results showed: The best time of testing vitality is 5 days after germinated. The germination rate was about 50% on the medium with 3% sucrose. Adding 60mg/L-100mg/L boric acid and 100mg/L Ca^2+ in the culture medium were benefit for pollen germination of Ginkgo biloba. The culture medium approach is as same accurate for pollen vitality determination as TIC. The best storage method is storing in 4℃ refrigerator.
