摘 要:研究苯并噻二唑(BTH)诱导苹果抗斑点落叶病的组织病理学特征,及其诱导抗病性与苯丙氨酸解氨酶活性(PAL)、几丁质酶活性和β-1,3-葡聚糖酶活性的关系。用整叶组织透明染色法进行组织病理学观察,用比色法测定PAL、几丁质酶和β-1,3-葡聚糖酶活的变化。结果表明:BTH能诱导苹果提高对斑点落叶病的抗性,接种前6d处理时可引起苹果斑点落叶病病情指数由对照的21.3下降到9.3。组织病理学观察发现,BTH诱导抑制病菌侵染钉的形成,限制病菌向细胞内扩展;酶活测定发现,BTH诱导能提高苹果叶片PAL、几丁质酶和β-1,3-葡聚糖酶活性。处理的果苗比对照果苗防卫反应启动的要早。Histopathological mechanism of apple leaves induced by benzothiadiazole (BTH) against Alternaira alternate f.sp.mali was researched. The relationship between BTH induced resistence and the activity of enzymes, including phenylalanine ammonialyase (PAL) , chitinase and β-1,3-glucanase , was studied. The histopathological response was investigated with whole leaf clearing and staining technique. The activity of enzyme was measured with colorimetric method. The result showed that BTH treatment could enhance the resistance of apples to Alternaira alternate f.sp.ma/i. When the leaves were treated at 6 days before inoculation, it could reduce the disease index from 21.3 to 9.3. The penetration pegs' formation was inhibited and pathogen extending was restricted. Activities of PAL, chitinase and β1,3-glucanase Increased. So the defendable reaction of induced apple trees was started earlier than control.
