摘　　要：目的通过比较S180肿瘤细胞体内、体外TLRs表达的变化,探讨TLRs在肿瘤发生、发展中的作用和意义。方法①将传代培养的S180腹水癌细胞接种于ICR小鼠腹腔左侧,建立S180腹水癌模型;②采用密度梯度离心法分离纯净肿瘤细胞;③采用半定量RT-PCR方法检测S180腹水癌体外传代培养细胞与荷瘤鼠体内瘤细胞TLR1-9 mRNA表达水平。结果TLR1、2、4、6在S180腹水癌体外传代培养细胞中表达水平高于荷瘤鼠体内瘤细胞;TLR7体外传代培养细胞中表达水平低于荷瘤鼠体内瘤细胞;TLR5、9体外传代培养细胞中有表达,荷瘤鼠体内瘤细胞无表达;TLR3、8体外传代培养细胞与荷瘤鼠体内瘤细胞均无表达。结论S180肿瘤细胞可表达TLRs,并且不同的TLRs在体内、体外的表达有所差异,提示TLRs在肿瘤的免疫逃逸中发挥一定的作用。Objective To study the difference between TLRs expressed on the surface of S180 cells in vitro and in vivo,and investigate the significance of TLRs in the genesis and development of tumor.Methods ①The ascitic S180 models were established by injected subcutaneously to the left abdominal of ICR mice with subculturing ascitic S180 cells.②Separate pure tumor cell with the method of density gradient centrifugation.③The expression of TLR1-9 mRNA were analyzed by semiquantitative RT-PCR.Results TLR1,2,4,6 expressed more in vitro than in vivo;TLR7 expressed more in vivo;TLR5,9 only expresses in vitro;TLR3,8 neither expressed in vitro nor in vivo.Conclusion TLRs may express on the surface of S180 cells.And the expressions are different between the cells which grew in vitro and in vivo.It suggests that TLRs may play roles in the tumor evasion.
