摘 要:目的:研究氯离子通道CLC-7在小鼠牙胚发育过程中的表达及分布,探讨其在牙齿发育过程中的作用。方法:提取小鼠牙胚中的总RNA并制备不同发育阶段的小鼠牙胚标本,应用RT-PCR和免疫荧光染色方法检测CLC-7在小鼠牙胚中的表达及分布。结果:RT-PCR检测发现新生小鼠牙胚有Clcn7mRNA的表达。免疫荧光染色发现E18小鼠牙胚开始有CLC-7的表达,CLC-7存在于牙胚多种细胞,如内釉上皮层细胞、中间层细胞、星网状层细胞、牙乳头细胞、成釉细胞、成牙本质细胞。结论:CLC-7在小鼠牙胚中呈特异性空间分布,可能参与牙胚发育过程的调节。Objective. To determine the expression and distribution of CLC-7 during the mouse tooth development and explore the possible function of CLC-7 in tooth formation, nethods:RT-PCR and immunofluorescence methods were used to investigate mRNA and protein expression of C1C-7 in the mouse tooth germ. Results : Clcn7 mRNA was observed in the tooth germ of new born mouse. Immunostaining showed that CLC-7 located in outer enamel epithelium cells, inner enamel epithelium cells, stellate reticulum cells, stratum intermedium cells, dental papillary cells, ameloblasts and odontoblasts in E l8 tooth germ (bell stage). Conclusion: CLC-7 has a spatial and temporal distribution during mouse tooth development and might contribute to the mouse tooth development.
