摘  要:目的研究鬼臼毒素纳米脂质体(PDP-SLN)对宫颈癌细胞系Hela细胞增殖及凋亡的影响。方法不同浓度(0.0005~5.000μmol/L)鬼臼毒素(PDP)及PDP-SLN分别处理Hela细胞,MTT法检测细胞增殖活性。Annexin V/PI染色,流式细胞术检测细胞凋亡。结果(1)PDP-SLN及PDP对Hela细胞增殖的抑制作用为明显的剂量依赖性和时间依赖性,同药物浓度同时间孵育条件下,PDP-SLN对细胞增殖的抑制效果明显优于PDP。24、48、72h的半数抑制浓度(IC50)PDP-SLN为4.10、0.65、0.20μmol/L,PDP为9.2、4.0、1.3μmol/L。(2)PDP-SLN及PDP均可以明显促进细胞凋亡。PDP-SLN24、72h诱导的凋亡率为90.8%和94.2%;PDP24、72h诱导凋亡率为64.1%和68.4%。结论PDP-SLN具有更强的增殖抑制和凋亡诱导效果。Objective To evaluate the anti-proliferative and apoptosis-inducing effect of podophyllotoxin solid lipid nanoparticles (PDP-SLN) in human cervical carcinoma cells in vitro. Methods Hela cells were treated with PDP and PDP-SLN at different concentrations (0.0005-5 μmol/L), and the proliferation of the cells was assessed using MTT assay and the apoptotic index was determined by flow cytometry. Results Both PDP and PDP-SLN showed obvious inhibitory effect on the cell proliferation in a dose- and time-dependent manner. At the same concentration, PDP-SLN produced stronger inhibitory effect on the cells than PDP, with IC50 24, 48, and 72 h after the cell exposure to PDP-SLN and PDP of 4.10, 0.65, 0.20 μmol/L and 9.2, 4.0, 1.3 μmol/L, respectively. Both PDP and PDP-SLN significantly induced the apoptosis of the Hela cell, and the apoptosis rates of the cells incubated in the presence of 0.5 μmol/L PDP-SLN reached 90.8% at 24 h and 94.2% at 72 h, significantly higher than the rate of cells incubated with PDP (64.1% at 24 h and 68.4% at 72 h, P〈0.01). Conclusion PDP-SLN can effectively suppress the proliferation and induce apoptosis of Hela cells in vitro.
