摘 要:目的方法利用液体芯片-飞行质谱技术进行血清中胶质瘤疾病相关蛋白质的筛选。结果将78例胶质瘤患者和68例正常对照者血清样本各随机分为模型建立组和模型验证组;应用液体芯片-飞行质谱技术测定模型建立组样本,进行了差异蛋白的筛选和诊断模型的建立;然后用模型验证组样本进行了差异蛋白和模型的验证;并对其中的蛋白质进行鉴定。经模型建立组胶质瘤患者与正常对照者的血清蛋白质图谱比较,得到差异蛋白17个并建立了诊断模型,建模蛋白质为1460(m/z)、1612(m/z)、1776(m/z)、1862(m/z)、1941(m/z)、3952(m/z)、4052(m/z)。模型敏感性为85%,特异性为97.06%。其中差异蛋白质1460(m/z)、1612(m/z)、2551(m/z)为肿瘤高表达,差异蛋白1941(m/z)为肿瘤低表达,经鉴定蛋白质1941(m/z)为高分子量激肽原。结论利用液体芯片-飞行质谱技术进行血清中胶质瘤疾病相关蛋白质的研究,得到了差异蛋白和诊断模型,为临床胶质瘤的诊断提供了新的思路;差异蛋白质的鉴定结果可能为胶质瘤的治疗提供有价值的途径。Objective To screen glioma' s disease-related proteins in serum by liquid chip-MALDI TOF Mass Spectrometry.Methods The serum samples of 78 glioma patients and 68 normal control subjects were randomly divided into two groups to build up the group of establishing model and the group of verifying, model.The serum samples were detected by liquid chip-MALDI TOF Mass Spectrometry. The samples of brain tumor and normal groups of establishing model were detected to screen the differential proteins and establish the diagnostic model. The differential proteins and the model were verified by the groups of verifying model. The interested proteins were identified. Results The serum protein spectrums of the group of establishing model including glioma patients and normal control subjects were compared and got 17 differential proteins. Among them, 7 differential proteins were selected to estab- lish the diagnostic model of glioma,the m/z were 1 460,1 612,1 776,1 862,1 941,3 952,4 052 respectively. The sensitivity and specificity of the model was 85 % ,97.06% respectively. The up-regulated proteins were 1 460(m/z), 1 612(m/z) ,2 551 (m/z);The down-regulated protein was 1 941(m/z).The identified result of 1 941(m/z)was high molecular weight kininogen.Conclusion The differential proteins and diagnostic model were got by liquid chip-MALDI TOF Mass Spectrometry after the comparison of glioma' s and normal serums spectrum. The result provided new thought for diagnosis of ghoma in clinics. The identification of differential protein maybe provide valuable path to the therapy of glioma.
