摘  要:目的探讨大鼠行为学改变与急性坏死性胰腺炎(ANP)的关系以及鱼油干预对其的影响。方法将SD大鼠分为造模成功的ANP组(A组)和ANP鱼油干预组(F组)以及对照组(C组),每组8只。A组、F组在术中经十二指肠穿刺向胰胆管逆行注入牛磺胆酸钠,C组仅穿刺十二指肠而不注药;F组术后皮下注射鱼油脂肪乳,A组、C组术后皮下注射普通脂肪乳。观察各组大鼠胰腺组织病理变化及鼠尾悬挂试验(TST)静止时间,同时测定各组大鼠第5天的血清过氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活力和淀粉酶浓度。结果A组、F组TST静止时间各时间点均显著长于C组(P<0.01);自第2天起F组短于A组,其中第4、5天的差异有统计学意义(P<0.05或0.01)。F组的炎症细胞浸润程度显著低于A组(P<0.01)。A组SOD、GSH-Px活力显著低于C组(P<0.01),鱼油干预后,F组SOD、GSH-Px活力显著升高(P<0.01)。TST静止时间与病理学改变、SOD、GSH-Px活力具有相关性(P<0.01)。结论鱼油可显著改变ANP模型大鼠出现的行为异常,这可能是通过缓解ANP炎症后得到的间接结果。Objective To investigate the effect on behavioral character of rats with acute necrotic pancreatitis (ANP). Methods Twenty four male Sprague-Dawley (SD) rats were divided into three groups (8 in each ): in group A and group F ANP was induced by retrograde injection of 3.5% sodium taurocholate into biliopancreatic duct through duodenal wall; and sham operation was performed in rats of group C. Fish oil emulsion was injected subcutaneously in group F, while general fat emulsion was given in group A and group C. The immobility time in tail suspension test (TST) and the grade of the severity of pancreatitis were observed. Five days after experiment the activity of serum antioxidant enzymes (SOD, GSH-Px)and the concentration of serum amylase in each groups were measured respectively. Results The immobility time in TST of group A and F were significantly longer than group C (P〈0.01) from d2 on. The immobility time in TST of group F was shorter than that of group A, however, only on the d4 and d5 (P〈0.05 or 0.01). Histopathological examination demonstrated that the infiltration of the inflammatory cells in group F was significantly milder than group A (P〈0.01). The activity of serum glutathione peroxidase (GSH-Px) and superoxide dismutase (SOD) in group A was significantly lower than those in group C (P〈0.01), and treatment of fish oil increased the GSH-Px and SOD levels in group F significantly. The immobility time in TST was correlated with histopathological changes and serum GSH-Px and SOD activity. Conclusion Fish oil can affect the ANP rats' behavioral character, which might be associated with the improvement of pancreatic inflammatory.
