摘 要:目的观察大鼠全脑缺血/再灌注损伤后海马PPARγ mRNA表达的动态变化。方法采用夹闭双侧颈总动脉,颈总静脉抽血后回输建立大鼠全脑缺血/再灌注损伤模型。Morris水迷宫检测大鼠空间定向能力变化,HE染色观察海马病理组织学改变及RT-PCR法检测缺血再灌注后不同时间点PPARγ mRNA的表达变化。结果全脑缺血/再灌注损伤导致大鼠空间学习及记忆能力明显下降,海马神经元出现明显的核固缩和细胞丢失。PPARγ mRNA的表达先升高后降低,以缺血/再灌注后48h表达水平最高,30d后接近正常水平。结论全脑缺血/再灌注损伤大鼠海马组织中PPARγ mRNA的表达,在再灌注30d内明显增加,表达高峰在48h。To observe the changes of PPARγ mRNA caused by global cerebral ischemia/reperfusion in rats hippocampi. Methods The model of global brainischemia/reperfusion(I/R) was induced by bilateral common carotid arteries occlusion and vena cervicalis retransfusion. Morris water maze was used for assessing the changes of spatial orientation and memory functions. HE staining was used for evaluation of pathological histology changes of hippocampal neurons, and RT-PCR was performed to detect changes of the PPARγ mRNA expressions. Results The spatial learning and memory ability significantly decreased in I/R rats, hippocampal neurons showed obvious karyopycnosis and loss. The expression of PPARγ mRNAin I/R rat hippocampussignificantly increased and reached the peak at 48 h after I/R . The PPARγ mRNA expression level was still higher than that of sham operated group although there existed no significance between the groups at 30d after I/R. Conclusion Global cerebral I/R can result in significant increase of PPARγ mRNA expressionin rat hippocampus. The peak time is at 48h after I/R.
