两种神经源性痛大鼠模型的比较

摘  要:目的:比较慢性坐骨神经缩窄模型(CCI)和L5脊神经前根切断(L5VRT)两种神经病理性疼痛模型的效果。方法:健康雄性SD大鼠26只,鼠龄8周,体重180~220g,随机分为3组:L5VRT组(n=10),CCI组(n=10),假手术组(n=6)。各组分别在左后肢制作模型。以热板法缩爪潜伏期和50%缩爪阈值作为行为学指标进行观察。记录有无死亡、致残、跛行、自噬行为。结果:自术后第1天起各组总体之间缩爪潜伏期、机械法50%缩爪阈值均存在差异(P<0.05),且L5VRT组与对照组,CCI组与对照组比较,其缩爪潜伏期阈值、机械法50%缩爪阈值均有所下降(P<0.05)。L5VRT组下降程度低于CCI组(P<0.05)。CCI组致残1例,跛行3例,自噬1例。L5VRT、对照组未出现。结论:L5VRT模型是一种成功率高、稳定、更接近临床实际的神经病理性疼痛模型。Objective: To compare the efficacy of neuropathic pain models:chronic constriction injury of sciatic nerve(CCI) and lumber 5 ventral root transaction (L5 VRT) model. Methods: Twenty six male SD rats(weighted from 150 to 200 g)were randomly divided into three groups:group L5 VRT(n =10) ,group CCI (n =10), group sham(n =6).Each group was operated on the let hind limb of rat. The thermal withdraeal latency (TWL) and mechanical withdrawl threshold (MWT) of rats were used to evaluate its behaviour, record the number of death, mutilation,limb and autophagy. Results: The TWL and MWT in group L5 VRT and group CCI all decreased since the first day after operation. The difference in scores responded more rapidly to TWL and MWT was lower in group CCI than that in group L5 VRT.There were one mutilation ,three limb and one autophagy appeared in group CCI. Conclusion: The lumber 5 ventral root transaction (L5 VRT) model is a high achievement ratio stabilize model and more practicable than the previous.
