摘　　要：目的:通过RT-PCR及免疫组织化学染色检测小眼畸形相关转录因子(MITF)mRNA及MITF蛋白的表达来探讨透明细胞肉瘤(CCS)的组织发生与起源。方法:收集5例CCS冰冻新鲜组织标本,24例福尔马林固定石蜡包埋组织标本。RT-PCR方法检测5例冰冻组织标本MITF基因mRNA的表达;24例CCS石蜡组织标本采用过氧化物酶染色(SP)方法进行免疫组化染色,检测S-100、HMB-45、MITF蛋白的表达。管家基因GAPDH为内参照。结果:4例CCS冰冻组织标本中检测到GAPDH mRNA表达,100%(4/4)检测到MITF mRNA表达。24例CCS石蜡组织标本中S-100、HMB-45及MITF总阳性率分别为79.2%(19/24)、70.8%(17/24)、83.3%(20/24)。结论:CCS中MITF在基因和蛋白水平高表达为神经嵴组织起源学说提供了依据。Objective： To discuss the genesis of clear cll sarcoma （CCS）by detecting the expression of MITF mRNA and MITF protein. Methods： Tumor tissues from 5 cases of fresh frozen tissues and 24 cases of formalin-fixed,paraffin-embedded tumor tissues of CCS were collected. MITF transcripts of 5 cases of fresh frozen tissues were detected by RT-PCR.The expressions of S-100,HMB-45 and MITF of 24 cases of formalin-fixed,paraffin- embedded tumor tissues of CCS were detected by immunohistochemistry. Results： RT-PCR analysis for the GAPDH was detected in 4 of 5 cases tested. Melanocyte- specific splice form of the MITF transcript was positive in 4 of 4 cases （4/4）. Most cases showed positive immunostaining for S-100 protein （19/24）, HMB45 （17/24）, MITF （20/24）.Conelusion： These data confirm that CCS expresses the melanocyte-specific form of the MITF transcript, further supporting its genuine melanocytic differentiation theory to some extent.
