摘  要:目的检测肺鳞癌组织中p14ARF蛋白及E2F-1蛋白的表达,探讨二者在肺鳞癌发生和发展中的作用及临床应用价值。方法采用免疫组织化学方法检测56例肺鳞癌组织和13例对照组(来自于肺炎标本)中p14ARF蛋白和E2F-1蛋白的表达。结果①在肺鳞癌和对照组中p14ARF蛋白表达的阳性率分别为41.1%和76.9%,肺鳞癌组远低于对照组,差异有显著性(χ2=5.435,P=0.020)。②E2F-1蛋白在肺鳞癌和对照组中阳性表达率分别为55.4%和7.7%,肺鳞癌组远高于对照组,差异有显著性(χ2=9.639,P=0.002)。E2F-1蛋白在肺鳞癌组织中阳性表达率随组织分化程度的降低而升高;在淋巴结已转移组的阳性表达率显著高于淋巴结未转移组,差异有显著性(χ2=5.396,P=0.020)。③p14ARF蛋白和E2F-1蛋白表达呈负相关(rs=-0.359,P<0.05)。结论p14ARF蛋白和E2F-1蛋白的表达异常可能与肺鳞癌的发生发展有关,p14ARF蛋白和E2F-1蛋白的联合检测可能为诊断肺鳞癌以及临床判断组织恶性程度提供参考。Objective To detect the expression of p14^ARF and E2F-1 in human squamous cell carcinoma of lung and explore their role in the development of human squamous cell carcinoma ( SCC ) of lung and clinicopathological significance. Methods Thirteen cases of inflammation of the lung (the control group) and 56 cases SCC of lung were enrolled in this study, All specimens were affirmed by pathology, p14^ARF protein and E2F-1 protein were detected by immunohistochemical examination of paraffin-embedded sections, Results (1)The positive rate of p14^ARF protein in SCC and the control group were 41, 1% and 76, 9% ,respectively,the difference was significant (x^2 = 5,435,P = 0.020), (2)The positive rate of E2F-1 in SCC was 55, 4% ,which was significantly higher than 7. 7% in control group( x^2 = 9, 639 ,P = 0. 002). Expression of E2F-1 in SCC was closely related to tumor stage, age and lymph node metastasis(P 〈0. 05), (3)There was an inverse correlation between p14^ARF protein and E2F-1 protein ( rs = - 0, 359, P 〈 0. 05 ), Conclusion The down-regulation of p14^ARF expression and up-regulation of E2F-1 expression may contribute to the carcinogenesis and development of SCC of lung and can be a biological marker for diagnosis and judging malignancy degree of SCC of lung.
