摘  要:目的探讨核转录因子NF-κB(nuclear factor-kappaB)的激活及基质金属酶9(MMP-9)的表达在腹主动脉瘤形成中的作用。方法成年雄性WISTER大鼠24只,随机分为腹主动脉瘤组、正常对照组(每组各12只)。用弹力酶灌注法建立大鼠腹主动脉瘤模型,免疫组织化学分析NF-κB,MMP-9的蛋白表达,RT-PCR方法检测其mRNA的表达。结果腹主动脉瘤组可见明显炎症细胞浸润,NF-κB被激活,同时伴有MMP-9表达增高。正常对照组未见明显炎症细胞浸润,仅有少量NF-κB的表达。结论NF-κB的激活、MMP-9表达增加在腹主动脉瘤病变的发生与发展过程中起着重要作用。Objoetive To study the role of nuclear factor Kappa B ( NF -κB) and MMP -9 at the early phase of AAA. Methods Twenty-four Wistar rats were randomly divided into 2 equal groups (n = 12). An animal model of AAAs was established by perfusion of the aorta with elastase. Immunohistochemistry staining was used to detect the expression of MMP -9 and NF -κB protein. The expression of MMP -9 and NF-κB mRNA was detected by RT - PCR. Resuit Inflammatory infiltration occurred in case of AAA. NF -κB and MMP - 9 expression significantly increased in AAA. Inflammatory infiltration was not found in normal aortas. Conclusion NF -κB and MMP -9 expression significantly increases in AAA. NF -κB and MMP -9 are considered to play an important role in the pathogenesis of AAA.
