苯胺和伯氨喹对大鼠高铁血红蛋白和网织红细胞生成的影响

摘　　要：目的 :了解伯氨喹 (伯喹 )对大鼠高铁血红蛋白 ( MHb)生成和网织红细胞 ( ret)增生的影响 ,并与苯胺所致的大鼠急性溶血反应进行比较。方法 :SD大鼠经腹腔 ( ip) 1次注射 3%苯胺花生油溶液 1ml/ kg,于给药前和给药后 2 h、2 4 h、4 8h和 72 h分别取血测定 MHb和 ret;另 1组大鼠灌胃 ( ig)伯喹 4 0 mg/ kg· d× 7d,于给药前及首剂给药的次日起 ,逐日取血检测 MHb和 ret直至停药后第 5d,以后隔日血检至停药后 15d。以上 2个给药组均设相应的溶剂对照。另对 37只大鼠进行了 G6PD活力测定。结果 :苯胺组给药后 2 h,MHb即显著升高 ,2 4 h恢复正常 ;ret自给药后 2 4 h开始上升 ,至 72 h仍持续升高 ,表现为急性血管内溶血反应。伯喹组给药期间及停药后MHb未见明显变化 ;ret于给药的 d5起持续低于对照组 ,至停药后的 d3— d5反应性增高 ,以后逐渐恢复正常。大鼠的 G6PD活力为 4 2 .3± 11.1%。结论 :伯喹对大鼠血液系统的影响表现为抑制网织红细胞的增生 ,但不引起类似苯胺所致的急性溶血反应。AIM:To understand the toxic effect of primaquine and aniline on the formation of methemoglobin (MHb) and proliferation of reticulocyte (ret) . METHODS:Two groups of male Sprague- Dawley rats were each given intraperitoneally with 3% aniline peanut oil solu- tion at a single dose of 1 ml/ kg and intragastrically with primaquine 40 mg/ kg per day for 7 days,respectively. Another two groups receiving corresponding solvents were served as con- trols. Blood MHb level and retcountwere determined at 2 h,2 4h,48h and 72 h after medi- cation for aniline and every day for primaquine from the day of the administration of the sec- ond dose of drug till the 5 th day after treatment,and then every other day till the 1 5 th day. RESULTS:The mean percentage concentration of MHb and ret count in aniline group rats was significantly higher than those of the controls at 2 h and 2 4h after aniline administra- tion,indicating that the rats in the aniline group presented acute intravascular hemolytic re- action. No remarkabale changes were found in the mean percentage concentration of MHb in primaquine group rats after medication,while the mean ret count was significantly lower during and after treatment than those before medication. CONCL USION:In rats,pri- maquine could inhibitthe proliferation of reticulocytes,butdid notinduce any acute intravas- cular hemolytic reaction similar to those seen in aniline group rats.
