子宫内膜癌盆腔淋巴结CK_(19)-mRNA的表达及临床意义

摘　　要：目的研究应用逆转录聚合酶链反应方法（RT—PCR）检测CK19-mRNA在子宫内膜癌盆腔淋巴结中的表达及其临床意义。方法取2006年6月～2007年3月于潍坊市人民医院行手术治疗的20例子宫内膜癌患者的130枚盆腔淋巴结，采用RT—PCR方法检测其CK19-mRNA的表达，取子宫内膜癌癌组织做阳性对照，取5例良性病变的子宫组织及盆腔淋巴结10枚做阴性对照。结果CK19-mRNA在20例子宫内膜癌癌组织中均呈阳性表达，在子宫良性病变患者内膜组织及淋巴结中均未表达。20例子宫内膜癌患者术后病理学检测仅1例发现癌转移，而应用RT-PCR检测有7例CK19-mRNA呈阳性表达，即可证实有肿瘤细胞的微转移。结论通过对子宫内膜癌患者盆腔淋巴结中CK19-mRNA的检测，进一步确定其临床病理分期，更有利于判断预后，指导治疗。Objective To explore the expression of cytokeratin 19 （ CK19 ） in lymph nodes of patients with endometrial carcinoma with reversetranscription-polyeerase chain reaction （RT-PCR） method and its clinical siguificanee. Methods Collecting 20 patients who were surgically staged and treated for endometrial carcinoma in Weifang People＇s Hospital from June 2006 to March 2007. RT-PCR method was used to determine the expression of CK19-mRNA in 130 lymph nodes. At the same time, the endometrial carcinoma were used as positive control. Uterine tissue and 10 lymph nodes from 5 patients with benign endometrial disease were used as negative control. Results All the endometrial carcinoma tissue were positive expression of CK19-mRNA,and all the normal lymph nodes and uterine tissue were negative expression. Only the lymph from one patient had been found cancer metastasis with Iraditional histopathology method. Seven palients＇lymph nodes were positive expression with RT-PCR method. Conclusion The detection of CK19-mRNA can be used to guide physician to judge clinical stage,to assess Ihe prognosis and provide effective treatment to patients with endometrial carcinoma.
