摘  要:目的:STAT3(signal transducer and activator of tran-scription 3)蛋白在多种肿瘤组织或细胞中高表达,研究表明STAT3蛋白可能参与了肿瘤的发生与发展。本研究拟设计针对STAT3的高效反义核酸,研究STAT3反义核酸对人白血病细胞HL-60增殖及凋亡的影响。方法:人工合成STAT3硫代ASODN,通过脂质体转染HL-60细胞,倒置显微镜下观察细胞形态学变化、应用四唑盐(MTT)比色法转染前后对细胞增生的影响,细胞凋亡检测法(POD)观察细胞凋亡。结果:①STAT3 ASODN抑制HL-60细胞增殖呈时间和浓度依赖性;②STAT3 ASODN可诱导HL-60细胞的凋亡,凋亡指数呈浓度依赖性。结论:脂质体转染STAT3 A-SODN能够明显抑制HL-60细胞增殖,且能诱导细胞凋亡,表明STAT3可能成为肿瘤治疗新的分子靶位。Objective:Signal transducer and activator of transcription 3(STAT3) protein was found constitutively activated in a wide variety of human tumor tissues and cell lines.It may be involved in tumorigenesis and development.This study was aimed at effective antisense oligonucleotide targeting STAT3 mRNA to explore its effect on the proliferation and apoptosis of human leukemic cell line HL-60.Methods: Anti-STAT3 phosphorothioate ASODN was synthesized and transfected into HL-60 cells by liposome.Cell morphological transformation was observed with inverted microscope while the effect on cell proliferation was examined by MTT assay after transfection,and cell apoptosis was determined in situ cell apoptosis detection kit(POD).Results: STAT3 ASODN inhibited HL-60 cells proliferation in a dose and time dependent manner and reduced the cell apoptosis with dose dependent in the apoptotic index.Conclusion: STAT3 ASODN can effectively inhibit the proliferation of leukemic cell lines HL-60 cells and induce the cell apoptosis,which supports that STAT3 may be a potential molecular target for tumor therapy.
