摘 要:目的:观察兔角膜新生血管的生长及退化过程中,基质金属蛋白酶、血管内皮生长因子的表达。方法:将45只健康新西兰大白兔随机取5只,为正常对照组,余40只作角膜缝线,建立角膜新生血管动物模型,随机分为阳性对照组和200μg/ml的GM6001用药组,用免疫组化及图像分析来研究两组的VEGF、MMP-9的表达和变化,并进行微血管计数。结果:在术后不同时间点,GM6001用药组MMP-9平均积分光密度(MMP-9 IOD)、VEGF IOD均较阳性对照组明显减少,差异有显著意义(P<0.001);VEGF IOD、MMP-9 IOD、微血管计数间有高度相关性(r=0.9840,r=0.7260)。结论:VEGF、MMP-9表达与角膜新生血管的形成与退化平行。GM6001可抑制和延迟实验性CRNV的形成。Objective: To observe the expression of matrix metalloproteinase and Vascular Endothelial Growth during the develop and degenerate of neovascularization in the rabbit cornea. Methods: The model of cornea new vessel was induced by suturing cornea. 5rabbits (ten eyes) from forty- five rabbits were randomly used as normal control. The others received suturing cornea, which were randomly divided into positive control group and GM6001 group . Immunohistochemistry analysis, technique and computerizeimage analysis were used to determine the expression and changes of VEGF, MMP-9, accounting microvessels respectively. Results:At rhe different periods, MMP-9 IOD, VEGF IOD and microvessel in positive control group were significantly lower than that in GM600I control group (P〈0. 001), and then exisiting significant correlativity( r = 0. 9840, r = 0. 7260). Concluson: The expression of MMP-9 and VEGF are parallel with corneal neovascularization. GM6001 can prevent and delay the corneal neovascularization.
