摘 要:目的观察姜黄素对高脂血症大鼠血栓素B2(TXB2)、6-酮-前列腺素F1α(6-Keto-PGF1α)的影响,探讨其延缓动脉粥样硬化形成的可能机制。方法60只大鼠随机分为正常组、模型组、辛伐他汀组、姜黄素组。正常组喂饲普通饲料,其他3组喂饲高脂饲料,共4周。第5周开始,姜黄素组、辛伐他汀组分别灌服姜黄素0.571 g/(kg.d)、辛伐他汀5.71 mg/(kg.d);正常组和模型组灌以相应剂量的0.9%氯化钠注射液,共6周。至10周末,取静脉血检测TXB2、6-Keto-PGF1α的水平。结果模型组大鼠血浆TXB2含量明显升高(P<0.01),6-Keto-PGF1α含量显著降低(P<0.01)。辛伐他汀组、姜黄素组与模型组比较,血浆TXB2水平明显下降(P<0.01),血浆6-Keto-PGF1α水平明显上升(P<0.01,P<0.05);辛伐他汀组与姜黄素组比较,血浆TXB2水平无统计学意义(P>0.05),血浆6-Keto-PGF1α水平姜黄素组高于辛伐他汀组(P<0.01)。结论姜黄素可增加前列环素含量,减少缩血管物质TXA2的分泌,防止血栓形成,延缓动脉粥样硬化的形成。Objective To observe the effect of Jianghuangsu on Thromboxane B2 (TXB2) ,6 - Keto - PGF1α to investigate its probable mechanism of action, Methods 60 rats used in this investigation were randomly divided into four groups: 15 in normal group, 15 in the model group 15 in the simvastatin group and 15 in Jianghuangsu group. The normal control group was fed with normal diet, and other three experimental groups with high - fat diet from the first day of the experiment for four weeks. The drugs were administrated by gavage from the beginning of the fifth week. The Jianghuangsu group were administrated with Jianghuangsu (0. 571 g· kg^-1 · d6-1 ), simvastatin group were administrated with simvastatin (5.71 mg·kg^-1 ·d^-1 ). Normal saline was given to rats of normal control group and model group (0.571 g·kg^-1 ·d^-1 ). Each group was given gavage for six weeks. At the ending of the tenth week,to detect the level of TXB2 ,6- Keto- PGF1α . Results Compared with normal group, the TXB2 content in model group was significantly increased ( P 〈 0.01 ), and the 6 - Keto - PGF1α content was significantly decreased ( P 〈 0.01 ). Compared with model group, the TXB2 contents in two treatment groups were significantly decreased ( P 〈 0.01 ), and the 6 - Keto - PGF1α contents were signhqcantly decreased ( P 〈 0.05). In two treatment groups, the TXB2 contents had no difference ( P 〉 0.05), and 6 - Keto - PGF1α content of Jianghuangsu group was higher than that of slmvastatin group ( P 〈 0.01 ). Conclusion Jianghuangsu could have the effects of anticoagulation, preventing thrombus and antiplatelet aggregation, enhancing the activity of PGI2 and decreasing the secretion of vaso - excitor material TXB2 .
