摘 要:将紫外分光光度计与高效毛细管电泳两种测定熊果酸的方法进行比较分析。结果表明,高效毛细管电泳法简便、快速,准确度和灵敏度均明显高于紫外分光光度法,并建立了高效毛细管电泳(HPCE)法测定山茱萸中熊果酸含量的方法。该方法建立的条件为以15 mmol/L磷酸氢二钠(Na2HPO4)-15 mmol/L硼砂(Na2B4O7)(pH9.22)、含10%甲醇(v/v)的缓冲体系作为运行电解质溶液,检测波长214 nm,分离电压20.0 kV,可实现熊果酸分离检测。研究表明,熊果酸检测浓度在0.57~0.95 mg/mL范围内与峰面积积分值线性关系良好,其回归方程为:Y=6708X-2720.6(R2=0.9946),平均加样回收率为97.43%(RSD为3.90%)。本方法准确、可靠,可用于山茱萸药材的质量检定与控制。By contrast, the method of determination of u rsolic acid by high performance capillary Electrophoresis is more simplicial, rapid, accurate and sensitive than the ultraviolet spectrophotometry. To establish a method of high performance capillary electrophoresis (HPCE) for content determination of ursolic acid in Comus officinalis Sieb.et Zucc.. The ursolic acid was baseline separated under the optimum buffer condition of 15 mmol/L disodium hydrogen phosphate+15 mmol/L sodium borate(pH9.22)+10% carbinol(vA,) at 214 nm of the wavelengh and 20.0 kV of constant voltage. The detected concentration of ursolic acid showed a good linear relationship with the peak area score in the range of 0.57-0.95 mg/mL, and the curve was Y= 6708X-2720.6 (R2=0.9946). The average recovery rate was 97.43% (RSD=3.90% ). The present method is accurate and reliable, and it can be used for the quality control of Comus officinalis Sieb.et Zucc..
